Detection of Human Papillomavirus Genotypes by Pyrosequencing Method in Uterine Cervical Cancer Samples

Abstract
Objective: Human papillomavirus (HPV) is considered as the primary etiologic agent of cervical cancer. The detection of HPV genotypes are very important for the detection of vaccine strategy and epidemiological purpose. The pyrosequencing method is one of the new methods used for HPV genotypes. In this study, detection of HPV genotypes were aimed by pyrosequencing method in patients with uterine cervical cancer. Materials and Methods: For this study, 38 paraffin-embedded cervical cancer and ten cancer negative cervical samples were used. HPV DNA in samples were determined by real time-PCR. Then, HPV genotypes were determined from HPV DNA positive samples using pyrosequencing. Results: According to the results of real time-PCR, 35 (92%) of 38 samples were found as positive for HPV DNA. With pyrosequencing of 35 HPV DNA positive samples, it revealed that HPV 16 was detected in 26 (68.42%), HPV 18 in 2 (5.26%), HPV 16 and Africian subtype 1 super-infection in four samples (13.15%) while HPV 16 and HPV 18 super-infection was found in one (2.63%), HPV 16 Africian subtype 1 and HPV 16 Africian subtype 2 in one (2.63%) and finally HPV 16 and HPV 33 super-infection were detected in other one (2.63%). Ten cancer negative cervical samples were found as negative for HPV DNA. Conclusion: According to these results, HPV 16 were shown to be the most important genotypes. Identification of HPV genotypes with pyrosequencing is cheaper and easier as compared to other methods such as classic nucleotide sequencing method. Thus, we suggest to use pyrosequencing method for genotyping of HPV.

Amaç: Human papillonavirus (HPV) servikal kanserlerin primer etiyolojik ajanı olarak değerlendirilir. HPV genotiplerinin tespiti için kullanılan moleküler testler aşılama stratejisinin oluşturulmasında ve epidemiyolojik amaçla oldukça önemlidir. Pyrosequencing yöntemi HPV genotiplerinin belirlenmesinde kullanılan yeni bir yöntemdir. Bu çalışmada pyrosequencing yöntemi ile uterin servikal kanseri tanısı olan hastaların HPV genotiplerinin belirlenmesi amaçlandı. Gereç ve Yöntem: Çalışmada 38 parafin bloktaki servikal kanser ve on kanser negatif servikal doku örneği kullanıldı. Örneklerin HPV pozitifliği real time-PCR yöntemi ile tespit edildi. Daha sonra, HPV pozitif olarak tespit edilen örneklerden HPV genotipleri pyrosequencing yöntemi kullanılarak belirlendi. Bulgular: Real time-PCR sonucunda, 38 adet servikal kanser örneğinin 35 (%92)'inde HPV DNA pozitif saptandı. HPV DNA pozitif örneklerin 26 (%68.42)'sında HPV 16, iki (%5.26)'sinde ise HPV 18 tespit edildi. Bir (%2.6) hastada HPV 16 ile HPV 18'in, dört (%10.5) hastada HPV 16 ile HPV 16 Africian subtip 1'in, bir (%2.6) hastada HPV 16 ile HPV 33 ve bir (%2.6) hastada ise HPV 16 Africian subtip 1 ile HPV 16 Africian subtip 2'nin süper infeksiyonları belirlendi. On adet kanser negatif kontrol servikal doku örneklerinde ise HPV pozitifliği tespit edilmedi. Sonuç: Çalışma sonuçlarına göre HPV 16 infeksiyonlarının servikal kanserin en önemli etkeni olduğu gösterildi. Pyrosequencing yöntemi ile HPV genotiplerinin belirlenmesi diğer klasik DNA dizi analizi yöntemlerine göre kolay ve ucuzdur. Bu nedenle, HPV genotiplendirme için pyrosequencing yöntemini önermekteyiz.
