Investigation of the Effects of the Antidepressant Agomelatine on Rat Myometrial Contractions

Abstract

Objective: Investigating of the possible effects of melatonin analog, agomelatine, on spontaneous and oxytocin-induced contractility of rat myometrium in vitro. Materials and Methods: In this study myometrium strips were removed from female SpraqueDawley rats in dioestrus and placed in a isolated tissue bath containing Krebs solution. Effects of agomelatine (50 ?M, 100 ?M and 200 ?M concentrations) on spontaneous and oxytocin-induced miyometrium contractions were studied. Luzindole, the melatonin receptor antagonist, was also used before the application of the agomelatine to determine whether action of agomelatine was melatonin receptor-mediated. Results: Agomelatine inhibited spontaneous and oxytocin-induced contractions of rats myometrium in a dose-dependent manner. After application of luzindole, inhibition was not reversed. Conclusions: In this study the inhibitor effect of agomelatine on spontaneous and oxytocininduced uterine contractility in rats was shown for the first time. This inhibitor effect of agomelatine may provide an advantage in inhibiting uterine contractions in pregnant women with abortion risk. After administration of the melatonin receptor antagonist luzindol, the inhibition does not reverse, indicating that agomelatine was not bound to melatonin receptors. These results support that agomelatin may have a potential to modulate myometrial function.

Amaç: Melatonin analoğu olan agomelatinin sıçan miyometriyumunda in vitro spontan ve oksitosin indüklü kasılmalar üzerindeki muhtemel etkisini incelemektir. Gereç ve Yöntem: Çalışmada diöstrustaki Spraque-Dawley cinsi dişi sıçanlardan alınan miyometriyumlar, içerisinde krebs solüsyonu bulunan izole organ banyosuna yerleştirildi. 50 ?M, 100 ?M ve 200 ?M konsantrasyonlarda uygulanan agomelatinin spontan ve oksitosin indüklü miyometriyum kasılmaları üzerindeki etkisi incelendi. Ayrıca agomelatinin melatonin reseptör aracılı etki edip etmediğini belirlemek için melatonin reseptör antagonisti olan luzindol, agomelatin uygulanmadan önce kullanıldı. Bulgular: Agomelatin, spontan ve oksitosin indüklü miyometriyum kasılmalarını doz bağımlı olarak inhibe etti. Luzindol uygulandıktan sonra ise inhibisyonun geriye dönmediği görüldü. Sonuç: Bu çalışmada, agomelatinin sıçanlarda spontan ve oksitosin indüklü uterus kontraktilitesi üzerindeki inhibitör etkisi ilk kez gösterildi. Agomelatinin bu inhibitör etkisi düşük riski taşıyan gebelerde uterus kasılmalarını inhibe etmek amacıyla kullanılmasında bir avantaj sağlayabilir. Melatonin reseptör antagonisti olan luzindol uygulandıktan sonra ise inhibisyonun geriye dönmemesi, agomelatinin melatonin reseptörlerine bağlanmadığını göstermektedir. Bu sonuçlar agomelatinin miyometrial fonksiyonu düzenleme potansiyeli olabileceğini desteklemektedir.
