Quantification of Lactobacillus spp in Yoghurt Marketing by Quantitative Real-time PCR in Şanlıurfa Province

Abstract
It has already known that the health and biological effects of yoghurt is based on the bacteria with probiotic efficacy which are responsible for the traditional milk fermentation. Thus, any possible increase or decrease on the amount of such bacteria is very critical in terms of human health. While the importance of the probiotics consumption is rising crucially, the microbiological quality detection of the yoghurt based on the probiotics should be considered carefully regarding the public health. We aimed to quantity the Lactobacillus spp. (LAB), recently reported as an important probiotic on the DNA molecules directly isolated from handmade yoghurt products marketing by local people in Şanlıurfa province and nearby cities with qPCR method. For the identification of the Lactobacillus spp. (LAB) from the DNA molecules isolated from the yoghurt products, specific primers for Lactobacillus spp. 16S-23S ribosomal RNA Intergenic Spacer Region (ISR) were utilized. The Ct range of qPCR analysis were detected between 14-32. In order to maintain the sensitivity of the quantitative analysis, a standard curve was plotted between 102 and 108 CFU/g by using a specific plasmid standard series. Thus we managed to determine a sensitive threshold of 102 CFU/g. Our results indicated that the amount of such important probiotic bacteria as Lactobacillus spp. was at lower levels in handmade yoghurt products offered in our region. The main reason of these levels can be caused from the maturation process of the product. The factors which are crucial for the production period and method should be considered vigilantly. Furthermore; instead of traditional microbiological methods, a novel real-time PCR method was used in Lactobacillus spp. quantification that have important probiotic role with regards to public health. Additionally it is confirmed that this rapid, sensitive and robust method can be employed for the identification
Özet
Yoğurt; geleneksel süt fermentasyonunda büyük bir rol oynayan ve probiyotik etkinlikleri olan bakterileri içermesi nedeniyle hayatımıza sağlık katan en önemli besin maddelerinden biridir. Günümüzde oldukça fazla tüketilen probiyotiklerin; yoğurtlardaki miktarlarının artması veya azalması ve mikrobiyolojik kalitelerinin tespiti halk sağlığı açısından irdelenmesi gerekmektedir. Bu çalışmada, Şanlıurfa ili ve çevre ilçelerinde satışa sunulan yoğurtlarda; probiyotik olarak önemi sıkça bildirilen Lactobacillus spp.'lerin (LAB) qPCR metodu ile tanımlanarak, kantitatif olarak miktar analizinin yapılması amaçlanmıştır. Yoğurt örneklerinden direk izolasyon ile elde edilen DNA'lardan Lactobacillus spp.'nin qPCR ile tespiti için; Lactobacillus spp. 16S-23S ribosomal RNA Intergenic Spacer Region (ISR) gen bölgesine spesifik primerler kullanılmıştır. Real-time PCR ile yapılan analizler sonucunda, 14 ile 32 aralığında Ct saptanmıştır ve kantitatif analizin hassasiyetini korumak için, miktarları belli plazmid standart kullanarak 102 ile 108 CFU/g arasında standart eğri elde edildi ve real-time PCR analizlerinde kantitasyon bu eğri ile gerçekleştirilmiştir. Bu sayede alt limit olarak 102 CFU/g gibi hassas bir alt limit belirlenmiştir. Sonuçlar, Lactobacillus spp. insan sağlığı açısından değerli, probiyotik bakterilerin, bölgemizde satışa sunulan ev yapımı yoğurtlarda daha düşük düzeyde bulunduğunu, bölgemizdeki bu yoğurtların, tüketime olgunlaşma süreci tamamlanmadan sunuluyor olabileceğini, bu ürünlerin üretim sürecinde dikkat edilmesi gerektiği tespit edilmiştir. Ayrıca sonuçlar, ülkemizde ilk kez klasik mikrobiyolojik yöntemler yerine, real-time PCR yönteminin insan sağlığı açısından probiyotik önemleri olan Lactobacillus spp.'lerin miktar analizlerinde hızlı, hassas ve güvenilir olarak tespiti için kullanılabilecek bir yöntem olduğunu göstermiştir
