Combined effects of X-rays and trimethoprim on the mitotic activity in hymphocyte cultures

Abstract

Purpose: This study investigates the damaging effects of X-rays on the structure of biomolecules in the cell and on the mitotic activity of the cells combined with the effects of trimethoprim (TMP) on mitotic activity in lymphocyte cultures. TMP interferes with the biosynthesis of monomers such as DNA, RNA and proteins. The biological effects of X rays are twofold: the direct effect of X ray is to deform the structure of biomolecules they collide with by breaking off the electrons. The indirect effect is that -while passing through the tissues, X ray loses energy and this energy is absorbed by the tissue. As a result a deformation in the structure of the biomolecules occurs. Structural deformation in biomolecules such as DNA, RNA, proteins, and enzymes have profound effects on the proper functioning of the organisms because they are crucial for the survival of the organism. Trimethoprim is a chemotherapeutic agent which is widely used in the treatment of infections. Material and methods: The blood taken from the nonsmoking radiodiagnostic unit technicians was used in the preparation of the lymphocyte cultures as time-dosage combinations. Results: In the statistical analysis, while the ratio of mitotic index decreased with increasing doses of TMP (p0.05). The usage of TMP with constant time and increased dosage decreased the ratio of mitotic index (p0.05). Conclusion: Trimethoprim application causes a decrease of mitotic index and this is inversely proportional with the dosage.

Özet

Amaç: Bu çalışmada X ışınının hücre biyomolekül yapısına ve mitotik aktiviteye etkisi ile çeşitli enfeksiyon hastalıklarının sağaltımı amacıyla kullanılan ancak mitoz mekanizmasında aksamalara yol açma riski bulunan trimethoprimin(TMP) lenfosit kültürlerinde mitotik aktiviteye kombine etkileri araştırılmıştır. X ışınının biolojik etkisi iki yönlüdür. Doğrudan etkisi çarptıkları biomoleküllerin yapısını deforme ederek elektron koparmak şeklindedir. İndirekt etkisi de dokudan geçerken absorbe olan X ışınının biomolekül yapılarında deformasyon oluşturması şeklindedir. DNA, RNA, protein ve enzim gibi biomoleküllerin yapısal deformasyonunun organizmanın düzenli çalışması üzerine etkisi büyüktür, çünkü bunlar organizma için yaşamsal önem taşır. Gereç ve yöntem: Çalışma beş bireyde yapıldı ve her birey için üç kontrol grubu kullanıldı. Kan örneklerinden mikrokültür yöntemi ile 72 saatlik lenfosit kültürleri hazırlandı ve üç farklı sürede TMP uygulandıktan sonra mitotik indeks incelendi. Bulgular: Yapılan istatistiksel değerlendirmede, artan TMP dozuna paralel olarak mitotik indeks oranı düşerken (P0.05). Trimethoprim uygulama süresi sabit tutularak-dozunun arttırılması mitotik indeks oranını düşürürken (P0.05). Sonuç: Trimethoprim uygulaması dozla orantılı olarak mitotik indekste düşmeye yol açmaktadır.
