Oxidative and antioxidative activity in diabetic retinopathy

Abstract
In this study, erythrocyte superoxide dismutase (SOD),glutathione peroxidase (GPx) activities as antioxidant enzymes and plasma lipid fractions as well as levels of lipid peroxides (LPO) were determined in patients with diabetic retinopathy. Although plasma levels of total cholesterol and triglycerides in the patient group were higher and HDL-cholesterol levels lower than those of control group, all of these changes were statistically significant. While plasma LPO levels were significantly higher in diabetic retinopathy group, erythrocyte GPx and SOD activities were significantly lower in the same group. In diabetes mellitus, non-enzymatically increased glycosylation may alter the activities of antioxidant enzymes. Due to decreased antioxidant capacity, the red cells of diabetic patients may become susceptible to oxidative stress. The damage caused by the complications of diabetes mellitus increases the formation of free radicals and the complications.Increased radicals result in the consumption and decrease of antioxidant stocks. Our results suggested that free radical damage is the most important factor in the pathogenesis of diabetic retinopathy and that a positive feed back interaction occurs between free radical damage and diabetic retinopathy.

Bu çalışmada, diabetik retinopatili hastalarda plazma lipit fraksiyonları ile lipit peroksit düzeyleri (LPO) ve antioksidan enzimlerden eritrosit glutatyon peroksidaz (GPx) ve süper oksit dismutaz (SOD) aktiviteleri tayin edilmiştir.Total kolesterol, VLDL-kolesterol, trigliserit düzeyleri hasta grubunda, kontrol grubundan daha yüksek; HDL-kolesterol düzeyleri ise kontrol grubundan düşük ve istatistiksel yönden anlamlı bulunmuştur. Plazma LPO düzeyleri diabetik retinopati grubunda yüksek, eritrosit GPx ve SOD aktiviteleri ise aynı grupta daha düşük bulunmuştur.Diabetde,non-enzimatik glikolizasyondaki artış antioksidan enzim aktivitesinde değişiklik oluşturur. Diabetik hastaların eritrositlerinin oksidatif strese hassas olmaları antioksidan kapasitede azalma meydana getirir. Diabetin komplikasyonları sonucu oluşan hasar radikal oluşumunu arttırırken oluşan radikaller sonucunda komplikasyonların artışı gerçekleşir. Artmış radikal oluşumu antioksidan enzim harcanması sonucu antioksidan stoklarını tüketerek azalmaya yol açar. Bu sonuçlar serbest radikal hasarının diabetik retinopati oluşumunda çok önemli bir faktör olduğunu ve serbest radikal hasarı ile diabetik retinopati arasında pozitif "feed back" bir etkileşim olduğunu düşündürmektedir.
