Oxidative stress and osmotic fragility in experimental hypothyroidism

Abstract

Aim: The aim of this study is to investigate lipid peroxidation, Superoxide dismutase (SOD) enzyme activity, glutathione (GSH) levels, and erythrocyte osmotic fragility in experimental hypothyroidism in rats. Material and Methods: Spraque Dawley type female rats that their weights were between 160 g and 200 g were used. For such a purpose rats were divided into two as control (n= 10) and hypothyroid (n=10) groups. During experimental period while both group animals were/ fed by standard food, methimazole (75mg/1OOg food) was given to the hypothyroid group by adding their food for 30 days. At the end of experiment, blood samples were taken from abdominal aorta of animals under ether anesthesia. As a last product of lipid peroxidation malondialdehyde (MDA) levels, SOD activity, GSH levels and osmotic fragility were measured in the blood. Results: MDA levels were determined 5.59 ± 0.86, 6.14 ± 0.95nmol/mL, SOD activity were assessed 2173.0 ± 316.1, 1992.8 ± 272.8U/mL and GSH levels were found to be 25.33 ± 2.91, 23.33 ± 3.46 mmol/L in control and hypothyroid groups respectively. When the osmotic fragility results were examined, osmotic fragility levels were found to be minimal 0.60 % NaCI and, maximal 0.32 % NaCl in control group, whereas erythrocyte osmotic fragility levels were determined minimal 0.56% NaCl, and maximal 0.36 % NaCl in hypothyroid group. When the hemolytic increment curve were examined, maximal hémolyse level was found to be 0.40 % NaCl in control group and 0.48 % NaCl in hypothyroid group. Conclusion: In our research that was studied under the hypothyroid condition, it has been observed that oxidative stress was not found to be effective on blood but erythrocyte osmotic fragility was significantly increased.

Amaç: Çalışmamızda deneysel hipotiroidide lipid peroksidasyonu , süperoksit dismutaz (SOD) enzim aktivitesi ve glutatyon (GSH) düzeyleri ile bağlantılı olarak eritrositlerin osmotik frajilitelerini incelemeyi amaçladık. Materyal-Metod: Çalışmada ağırlıkları 160-200 g arasında Spraque Dawley türü dişi sıçanlar kullanıldı. Kontrol (n=10) ve hipotiroidi (n=10) olmak üzere iki grup oluşturuldu. Kontrol grubu standart yem ile beslenirken hipotiroidi grubuna yaklaşık 30 gün süre ile methimazol (75mg / 100 g yem) verildi. Deney süresi sonunda eter anestezisi altında sıçanların abdominal aortalarından kan örnekleri alındı. Kan örneklerinde lipid peroksidasyon göstergesi olarak malondialdehit (MDA) düzeyleri, SOD aktiviteleri; GSH düzeyleri ve eritrositlerin osmotik frajiliteleri ölçüldü. Bulgular: Çalışmamızda kontrol ve hipotiroidi grubunun MDA düzeyleri sırasıyla 5.59±0.86, 6.14±0.95 (nmol/ml), SOD aktivitesi 2173.0±316.1, 1992.8±272.79 (U/mL), GSH düzeyleri ise 25.33±2.91, 23.33±3.46 ($mu$M) olarak bulundu.Eritrosit osmotik frajilitesiyle ilgili osmotik direnç sınırları kontrol grubunda minimal % 0.60 NaCl, maksimal % 0.32 NaCl olarak bulundu. Hipotiroidi grubunda ise minimal osmotik direnç sınırı %0.56 NaCI, maksimal osmotik direnç sınırı % 0.36 NaCI olarak saptandı. Hemolitik inkrement eğrisi incelenerek saptanan maksimal hemoliz, kontrol grubunda %0.40 NaCl, hipotiroidi grubunda ise % 0.48 NaCl olarak belirlendi. Sonuç: Çalışmamızda, hipotiroidi koşulunda oksidatif streste herhangi bir değişiklik bulunmamasına rağmen eritrosit osmotik direncinin azaldığı gözlenmiştir.
