The effect of different in vitro conditions to in vivo development of Plasmodium berghei

Abstract
In this study, the effect of different in vitro conditions to in vivo development of Plasmodium berghei was investigated.We used two stains of P. berghei to infect mice and rats; (1) P. berghei obtained from National Institute of Prophylactic Medicine in Bakü-Azerbaycan, and (2) P. berghei kept in the cryobank of Immunology Department. In the first experiment, we injected 22 mice and 6 rats with the first strain of P. berghei and found atypical forms of parasite in the blood of mice and rats after 3-4 days of post infection the parasites did not show any further development and disappeared on the following days. In the second experiment we injected 47 mice with the second strain of P. berghei and found the typical forms of parasite in the blood of mice after 3-4 days of post infection. The infected blood obtained from mice in the second experiment was used for the following experiments to show the effect of in vitro conditions to infectivity of P. berghei. (1) The infected blood was kept in +4° C for 6 hours then taken to 20-26° C for 24 hours. All of mice infected with this blood became ill. (2) The infected blood kept in 20-26° C for 4 hours then in +4° C for 24 hours was injected to the mice. Half of the mice injected with this blood became ill which means that the infectivity was 50%. (3) The infected blood kept in 22-28° C for 24 hours in RPMI-1640+Hepes medium was injected to the mice. Infectivity of this blood was 75%. When the blood was kept more then 24 hours (48 and 72 hours) in the same conditions parasites didn't show any development in mice. Infectivity was 0%. (4) The infected blood kept in 22-28° C for 24 hours in other RPMI-1640 medium without hepes was injected to the mice. Half of the mice injected with this blood became ill. Infectivity was 50%. When the same blood was kept 48 hours in the same conditions the parasites didn't show any development at all in mice. (5) The infected blood kept in 22-28° C for 24 and 48 hours in citrate environment injected to mice showed no infection.
Özet
Bu çalışmada, in vitro koşulların kemiricilerin sıtma etkeni olan Plasmodium berghei'nin in vivo gelişimine etkisi araştırıldı. Çalışmamızda iki ayrı suş kullanıldı; birisi Milli-Araştırma Tıbbi Profilaktika Enstitüsü (Bakü-Azerbaycan)'nden temin edilen, diğeri İmmünoloji Bilim Dalı kriyobankında bulunan P. berghei idi. Birinci suşla 22 fare ve 6 sıcan, ikinci suşla 47 fare enfekte edildi. İn vitro olarak Bakü'den getirilen P. berghei farelere enjekte edildikten 3-4 gün sonra mikroskobik muayene yapıldığında periferik kanda atipik parazitler görüldü ancak takip eden günlerde gelişme olmadı ve tamamen yok oldu. Kriyobankda tutulan P. berghei farelere enjekte edildikten 3-4 gün sonra tipik parazit formları kanda görüldü. Enfekte farelerden alınan kanlar çeşitli süre ve sıcaklıklarda in vitro bekletildi ve P. berghei'nin enfektivliği araştırıldı. Bunun için aşağıdaki denemeler yapıldı ve sonuçlar elde edildi: (1) Fareden alınan kan 6 saat +4° C de ve 24 saat 20-26° C de tutuldu ve daha sonra fareler bu kanla enfekte edildi. Bu farelerin tamamında parazitlerin geliştiği gözlendi. (2) Fareden alınan kan 4 saat 20-26° C de ve ilave olarak 24 saat +4° C de tutuldu ve daha sonra bu kanla enfekte edilen farelerin ancak %50'sinde parazitin geliştiği gözlendi. (3) RPMI-1640+Hepes ortamında 24 saat (22-28° C) bekletilen kan farelere enjekte edildiğinde farelerin %75'inde parazit gelişirken aynı ortamda 48 ve 72 saat bekletildikten sonra enfekte edilen farelerde gelişme olmadı. (4) RPMI-1640 ortamında 24 saat (22-28° C) bekletilen kan ile enfekte edilen farelerin ancak %50'sinde parazit gelişirken, aynı ortamda 48 saat bekletildikten sonra enfekte edilen farelerde ise hiç gelişme olmadı. (5) Sitratlı ortamda 22-28° C de 24 ve 48 saat bekletilen kanla enfekte edilen farelerin hiçbirinde parazit gelişmedi.
