The comparison of Elisa and PCR in diagnosis of Chlamydia trachomatis infections in sex workers

Abstract

This study was performed to compare the PCR (16S rRNA+cryptic plasmid) with direct ELISA in diagnosis of asymptomatic endocervical chlamydial infections in 68 sex workers. In our study, the PCR tests using primers that target 16S rRNA and primers that target the specific sequence of 215 base pair that belongs to plasmid and ELISA method were compared. ELISA method using specific rabbit monoclonal antibodies to MOMP protein of C. trachomatis was employed. Of the 68 endocervical specimens from sex workers, a total of 17 (25%) were positive for C.trachomatis by PCR; 13 (19%) endoservical specimens were positive for 16SrRNA+cryptic plasmid product and 4 (6%) were positive only for cryptic plasmid product. Whereas, positive results were obtained in 10 (14.7%) of the specimens using ELISA. These data indicate that PCR which targets cryptic plasmid is more sensitive (100%) than ELISA (58.8%) for detecting C.trachomatis from genital specimens.

Özet

Bu çalışma, 68 hayat kadınında asemptomatik olarak seyreden endoservikal Chlamydia trachomatis enfeksiyonlarının tanısında, PCR (16S rRNA+kriptik plasmit) ve direkt ELİSA testlerinin karşılaştırılması amacıyla yapıldı. Çalışmada 16S rRNA'i hedef alan primerlerin kullanıldığı PCR yöntemi ile kriptik plasmide ait 215 baz çiftlik spesifik diziyi hedef alan PCR, ELİSA yöntemi ile kıyaslandı. Endoservikal örneklerdeki Chlamydia antijeni C. trachomatis'in MOMP proteinine karşı hazırlanmış monoklonal antikorların kullanıldığı ELİSA testi ile araştırıldı. Hayat kadınlarından alınan 68 endoservikal örnekden PCR ile 13 (%19)'ünde 16S rRNA+kriptik plazmid'e ait ve 4(%6)'ünde sadece kriptik plazmide ait olmak üzere toplam 17 (%25)'sinde hedef diziler tespit edilirken, ELİSA yöntemi ile örneklerin 10 (%14.7)'unda pozitif sonuç elde edildi. C. trachomatis'in tanısında, kriptik plazmidi hedef alan PCR yönteminin (%100) ELİSA'dan daha duyarlı (%58.8) olduğu tespit edildi.
