The effects of truncal vagotomy on blood glucose in an experimental rat model of diabetes

Abstract

In this experimental study, the effects of fruncal vagotomy on post-prandial glucose levels and intravenous glucose tolerance test in the streptozocin-induced diabetes model of rats were studied. A total of fifty Swiss type Albino rats were randomly divided into five groups, each was having 10. In-group 1, following laparotomy, vagal nerves were isolated. In groups 2, 3 and 5, diabetes was created by intraperitoneal administration of streptozocin; then bilateral fruncal vagotomy wax performed in group 3 and in group 5, only the vagal nerves were isolated by laparotomy. The only procedure was bilateral truncal vagotomy in non-diabetic group 4. In rats with streptozocin-induced diabetes, post-prandial, glucose levels significantly decreased, following truncal vagotomy, while no difference was noted in glucose tolerance test. In diabetic rats, cessation of vagal stimuli decreased post-prandial glucose levels, however, we believe that, new experimental studies including the assessment of plasma insulin and haemoglobin A 1c levels and histopathological evaluation of pancreatic islet cells are needed for the thorough understanding of the mechanism.

Özet

Bu deneysel çalışmada, streptozocin ile deneysel diabetes mellitus oluşturulan sıçanlarda trunkal vagotominin postprandial kan glikozu ve intravenöz glikoz toleransı üzerine olan etkileri incelendi. Elli adet İsviçre türü beyaz sıçan onarlı beş gruba ayrıldı. Grup 1'deki sıçanlarda laparotomi ve nervus vagusların izole edilmesi (n=10), grup 2'de yalnızca streptozotocin ile diabet oluşturulması (n=10), grup 3'de diabet oluşturulduktan sonra bilateral trunkal vagotomi (n=10), grup 4'de sadece bilateral trunkal vagotomi (n=10), grup 5'de ise diabet oluşturulduktan sonra, laparotomi ve nervus vagusların izolasyonu uygulandı (n=10). Deneysel diabet oluşturulan sıçanlarda trunkal vagotomi, postprandial kan şekerlerinde anlamlı düşmeye neden olurken, glikoz toleransında değişiklik yapmadı. Diabetik sıçanlarda vagal uyarının ortadan kalkması postprandial kan glikoz değerlerinde düşüklüğe neden olurken bu etkinin hangi mekanizmalarla oluştuğunun saptanması için, plazma insülin, hemoglobin A 1c düzeylerinin ölçümü ve pankreas adacık hücrelerinin histopatolojik incelenmesini de içeren yeni çalışmalara ihtiyaç olduğu kanısındayız.
