Analysis of the pathogenicity level of the live infectious bursal disease (Gumboro) vaccines used in Turkey by a molecular marker

Abstract

In this study, a 743bp fragment of the VP2 gene of 23 live infectious bursal disease (Gumboro/IBD) vaccine viruses with different pathogenic strength (intermediate, intermediate plus /hot) that are imported and used in the field of Turkey, was amplified by Reverse Transcriptase/Polymerase Chain Reaction (RT/PCR) technique and analyzed by the use of NgoMIV molecular marker. In this study, the ability of NgoMIV molecular marker to distinguish the pathogenic viruses which have the alanine amino acid at position 284 in the VP2 gene, from the attenuated vaccine strains that do not contain this amino acid at the same position, is utilized. The 743bp band of thirteen attenuated vaccine strains that are known to be in intermediate strength were not digested with NgoMIV enzyme and remained uncut. Two bands of 507bp and 227 bp were formed in the 9 out of 10 vaccines viruses that are known for their hot/intermediate plus characteristics. 743bp band of Bursine plus vaccine which is reported to be an intermediate plus vaccine according to its pathogenic characteristics was not digested by NgoMIV enzyme. In this study, it was observed that 22 of the 23 Gumboro vaccines that were examined by NgoMIV marker analysis carried the pathogenic characteristics of those which were claimed in their protocol dossiers. The different result obtained with Bursine plus did not conflict with the literature.

Özet

Bu çalışmada, Türkiye’ye ithal edilerek sahada kullanılmakta olan farklı patojenik karakterdeki (intermediate, intermediate plus/hot) 23 adet canlı infeksiyöz bursal hastalığı (Gumboro/IBD) aşı virusunun VP2 geninin 743bp’lik bölümü reverse transcriptase/polymerase chain reaction (RT/PCR) tekniği ile çoğaltılarak NgoMIV moleküler marker’ı ile analiz edildi. Çalışmada, NgoMIV moleküler marker’ının VP2 geninin 284 pozisyonundaki alanine amino asitini içeren patojenik virusları, aynı noktada bu aminoasiti içermeyen attenue aşı suşlarından ayırt edebilme özelliğinden yararlanıldı. İntermediate (orta kuvvetli) sınıfta olduğu bilinen 13 attenue aşı suşunda 743 bp’lik bant NgoMIV enzimi ile kesilmeyerek korundu. Hot/intermediate plus (kuvvetli) sınıfında olduğu bilinen 10 adet aşı virusundan 9 adedinde 507bp ve 227 bp’lik iki bant oluştu. Patojenik özellikleri ile intermediate plus olduğu bildirilen Bursine Plus aşısına ait 743bp’lik bant NgoMIV enzimi ile kesilmedi. Bu çalışmada, NgoMIV marker analizi ile incelenen 23 adet Gumboro aşısından 22 adedinin protokol dosyasında bildirilen patojenik grubun özelliklerini taşıdığı görüldü. Bursine plus aşısına ait farklı sonucun literatür bilgileri ile çelişmediği görüldü.
