Molecular analysis of infectious bursal disease field and vaccine viruses for their antigenic and pathogenic characteristics

Abstract
In this study, 140 field samples that were collected in 3 different periods from the beginning of the1990’s, when theinfectious bursal disease (IBD) outbreaks started in Turkey, until 2010 (1990-1994; 1999-2003 and 2010) were analyzedcomparatively with the reference infectious bursal disease virus (IBDV) strains (STC, SAL, D78, IN, Ev, MD) and 20 live IBDVvaccines used in the field by Reverse Transcriptase/Polymerase Chain Reaction-Restriction Fragment Length Polymorphism(RT/PCR-RFLP) technique. In these analyses, a 743bp fragment of the VP2 gene of IBD viruses were amplified and antigenicgrouping of field, vaccine and reference viruses were investigated with BstNI(BstOI) and MboI enzymes, pathogeniccharacteristics were examined with SspI enzyme.IBDV vaccines used in Turkey were found to be in molecular groups 3, 4a, 4b, 5 and 6. Antigenic grouping of 72 fieldviruses that were positive in RT/PCR tests, isolates were detected in molecular groups 3, 4a and 6. Also, 16 new differentmolecular groups were observed. Most of the field viruses were clustered in molecular groups 6 and New 1. Other field isolateswere distributed among New 2 to New 16. No American variant type virus profiles (molecular group 1 and 2) were observed inthe field. The RFLP profile diversity obtained with the MboI enzyme was greater than the one obtained by BstNI (BstOI) enzymeamong the wild type field viruses. As a result of pathogenic grouping study with SspI enzyme, 65 of the 72 field isolates (90.27%) were found to be very virulent (vvIBDV), 6 (8.33 %) of them classical virulent in nature. One field isolate (E40) showed adifferent RFLP than others when analyzed by SspI enzyme.

Bu çalışmada, Türkiye’de infeksiyöz bursal hastalığı (IBD) salgınlarının başladığı 90’lı yılların başından 2010 yılına kadar3 farklı dönemde (1990-1994; 1998-2003 ve 2010) toplanan 140 saha izolatı, referans infeksiyöz bursal hastalığı virus (IBDV)suşları (STC, SAL, D78, IN, Ev, MD) ve sahada kullanılmakta olan 20 adet canlı IBDV aşısı ile karşılaştırmalı olarak ReverseTranscriptase/Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (RT/PCR-RFLP) tekniği ile incelendi. Buanalizlerde, IBD viruslarının VP2 geninin 743bp’lik bir bölümü amplifiye edilerek, saha, aşı ve referans virusların antijenikgrupları BstNI(BstOI) ve MboI enzimleri ile, patojenite özellikleri SspI enzimi ile araştırıldı.Türkiye’de kullanılan aşı viruslarının 3, 4a, 4b, 5 ve 6. moleküler gruplarda yer aldığı görüldü. Reverse Transcriptase/Polymerase Chain Reaction analizinde pozitif reaksiyon veren 72 saha virusunun antijenik gruplandırmasında; 3, 4a ve 6.moleküler gruplarda yer alan izolatlar tespit edildi, ayrıca yeni 16 farklı moleküler grup gözlendi. Saha viruslarınınçoğunluğunun 6 ve Yeni 1 moleküler gruplarında toplandığı görüldü. Diğer saha izolatları Yeni 2’den –Yeni 16’ya kadar olanmoleküler gruplar arasında dağılım gösterdi. Sahada Amerikan tipte antijenik varyant virusların RFLP profiline (Moleküler Grup1 ve 2) rastlanmadı. Saha virusları arasında MboI enzimi ile elde edilen RFLP profili çeşitliliği BstNI(BstOI) enzimi ile eldeedilene göre daha yüksekti. SspI enzimi ile patojenik gruplandırma çalışması sonucunda, incelenen 72 saha izolatının 65 adedinin(% 90.27) çok virulent (vvIBDV), 6 adedinin (% 8.33) klasik virulent yapıda olduğu görüldü. Bir saha izolatı (E40) SspI enzimiile diğerlerinden farklı RFLP oluşturdu.
