Determination of the effect of different drying temperatures on the content and chemical composition of essential oil of sage (Salvia officinalis)

Abstract

The aim of this experiment was to determinate the effect of different drying temperatures on the content andchemical composition of essential oil of sage (Salvia officinalis L.). Essential oil isolated for 3 hours from fresh and ovendried leaf samples (at 35°C, 45°C, 55°C and 65°C) by using Clevenger type apparatus was analyzed with GC-MS. Thecontent of volatile oils (%) in different drying temperatures was in the order of: fresh leaf sample (0,483%), dried leafsample at 35 °C (1,837%), at 45 °C (1,550%), at 55 °C (1,267%) and at 65 °C (1,263%). In total, 36 compounds ofessential oil were identified from fresh and dried leaf samples. The main compounds of essential oils isolated in differentdrying temperatures were α-thujone and camphore. α-Thujone was highest at fresh leaf sample (28,53%) and lowest at45 °C (11,21%). The highest and lowest camphore contents were observed at sample dried at 35 o C (32,53%) and freshleaf sample (23,62%). On the other hand, the ratio of 1,8-Cineole, effective in the medical use of sage, has also changeddepending on different drying temperatures. The highest and lowest 1,8-Cineole ratios were observed at sample dried at35 o C (9,480%) and fresh leaf sample (7,947%). Results showed that different drying temperatures were effective onessential oil content and chemical composition of essential oil of sage.

Özet

Bu araştırmanın amacı Tıbbi adaçayında (Salvia officinalis L.) farklı kurutma sıcaklıklarının uçucu yağ oranı ve uçucu yağın kimyasal kompozisyonu üzerine etkisini belirlemektir. Taze ve kurutulmuş yaprak örneklerinden (35°C, 45°C, 55°C ve 65°C de) üç saat süreyle clevenger cihazı kullanılarak çıkarılmış olan uçucu yağlar GC-MS cihazı ile analiz edilmiştir. Farklı kurutma sıcaklıklarından elde edilen uçucu yağ oranları (%) sırasıyla: % 0,483 (taze yaprak örneği), % 1,837 (35 °C), % 1,550 (45 °C), % 1,267 (55 °C) ve % 1,263 (65 °C)’dir. Taze ve kuru yaprak örneklerinden elde edilen uçucu yağlarda 36 farklı bileşen tespit edilmiştir. Farklı kurutma sıcaklıklarında elde edilen uçucu yağların ana bileşenleri α-thujone ve camphore’dur. α-thujone oranı taze yaprak örneklerinde en yüksek (% 28,53) iken, 45 °C de kurutulmuş yaprak örneklerinde en düşük (% 11,21) olarak bulunmuştur. En yüksek ve en düşük camphore oranları ise 35 o C’de kurutulmuş yaprak örneklerinde (% 32,53) ve taze yaprak örneklerinde (% 23,62) belirlenmiştir. Diğer taraftan, tıbbi adaçayının tedavi amaçlı kullanımında önemli olan 1,8-Sineol (ökaliptol) oranı aynı zamanda farklı kurutma sıcaklıklarına bağlı olarak değişiklik göstermiştir. 1,8-Sineol oranları 35 0 C de kurutulmuş örneklerde en yüksek (% 9,480) ve taze yaprak örneklerinde en düşük (%7,947) olarak gözlemlenmiştir. Sonuçlar; tıbbi adaçayında farklı kurutma sıcaklıklarının uçucu yağın oranı ve uçucu yağın komposizyonu üzerinde etkili olduğunu göstermektedir.
