Extracellular biosynthesis of silver nanoparticle using Tilia rubra DC. extract and antifungal activity

Abstract

Although nanoparticles synthesized by chemical reactions show a wide range of toxicities in various vertebratesand invertebrates, they are produced in large quantities to take advantage of their antifungal activities. Due to theremarkable disadvantages of conventional chemical methods, the green synthesis of metallic nanoparticles has acquiregreat attention in recent years. This method is proposed as an alternative to classical chemical methods with its low cost,environmentally friendly and simple approaches. Our aim in this study is to determine the suitability of Tilia rubra DC.extract in the extracellular synthesis of silver nanoparticles and characterize the material obtained. T. rubra extract hasbeen studied for the first time for the biosynthesis of silver nanoparticles, forming surface plasmon resonance at 427 nmin UV-Vis spectrophotometer. Also, transmission electron microscope (TEM) images showed that nanoparticles havespherical morphology. According to TEM images, nanoparticles were found to be between 5-15 nm in size. X-raydiffraction (XRD) analysis, on the other hand, determined that the particles were crystalline with a face centered cubicgeometry. Fourier transform infrared (FTIR) analysis was performed to identify possible biomolecules responsible forbio-reduction of silver ions. The antifungal effect of the biosynthesis silver nanoparticles on Candida albicans, apathogenic yeast, was examined by the agar diffusion method, and as a result, the synthesized silver nanoparticles showeda significant inhibitory effect on C. albicans. Our results support the potential for future use of biosynthesized silvernanoparticles for industrial and biomedical applications, thanks to the environmentally friendly synthesis procedure.

Kimyasal reaksiyonlarla sentezlenen nanopartiküller çeşitli omurgalı ve omurgasızlarda geniş bir toksisite yelpazesi sergileseler de, antifungal aktivitelerinden yararlanmak üzere büyük miktarlarda üretilmektedir. Klasik kimyasal yöntemlerin dikkat çekici dezavantajları nedeniyle, metalik nanopartiküllerin yeşil sentezi son yıllarda büyük ilgi görmeye başlamıştır. Bu yöntem düşük maliyetli, çevre dostu ve basit yaklaşımları ile klasik kimyasal yöntemlere bir alternatif olarak önerilmektedir. Bu çalışmada amacımız, gümüş nanopartiküllerin hücre dışı sentezinde Tilia rubra DC. ekstraktının uygunluğunu belirlemek ve elde edilen materyali karakterize etmektir. Bunun yanısıra biyosentezlenmiş gümüş nanopartiküllerinin antifungal etkisi değerlendirilmiştir. T. rubra ekstraktı, gümüş nanopartiküllerin biyosentezi için ilk kez çalışılmış olup UV-Vis spektrofotometresinde 427 nm'de yüzey plazmon rezonansı oluşturmuştur. Ayrıca geçirimli elektron mikroskop (TEM) görüntüleri nanopartiküllerin küresel morfolojiye sahip olduğunu göstermiştir. TEM görüntülerine göre nanopartiküllerin boyutlarının 5-15 nm arasında olduğu bulunmuştur. X-ışını kırınımı (XRD) analizi ise partiküllerin yüz merkezli kübik geometriye sahip kristal yapıda olduğunu belirlemiştir. Gümüş iyonlarının biyoindirgenmesinden sorumlu olası biyomolekülleri tanımlamak için Fourier dönüşümlü kızılötesi spektroskopisi (FTIR) analizi yapılmıştır. Biyosentezlenmiş gümüş nanopartiküllerin patojen bir maya olan Candida albicans üzerindeki antifungal etkisi agar difüzyon metodu ile incelenmiş ve sonuç olarak sentezlenen gümüş nanopartikülleri, C. albicans üzerinde önemli inhibe edici etki göstermiştir. Sonuçlarımız, biyosentezlenmiş gümüş nanopartiküllerin çevre dostu sentez prosedürü sayesinde, gelecekte endüstriyel ve biyomedikal uygulamalar için kullanım potansiyeli olduğunu desteklemektedir.
