The Effects of Nigella Sativa (Corek Otu) Seeds on Plasma Nitric Oxide, Malondialdehyde and Whole Blood Glutathione Concentrations in Geese

Abstract
In this study, the effects of Nigella sativa seeds on plasma malondialdehyde (MDA), nitric oxide (NO) and whole blood glutathione (GSH) concentrations were investigated in geese. For this purpose, 8 Anser anser breed geese were fed with a diet containing 5 % ground Nigella sativa seeds for 6 weeks. Blood samples were properly collected from V. subcutenea ulnaris at the beginning and every week of the study for 6 weeks. Biochemical analysis of plasma MDA and blood GSH concentrations were spectrophotometrically measured on a weekly basis according to the methods of Yoshioka et all. and Beutler et all. respectively. Plasma NO concentration was measured by the method of Miranda et all. with a microplate reader. Alterations in measured parameters were determined with respect to the weeks of study. While MDA concentration was significantly decreased at the 2nd week and continued to be decreased compared to 1st week, blood GSH level was significantly increased and remained high compared to 1st week of study. However, no alteration was found in plasma NO concentration among weeks. In conclusion, Nigella sativa administration for 6 weeks resulted in significant decrease in MDA levels and significant increase in GSH levels of geese implying that Nigella sativa could be protective against oxidative stress.
Özet
Bu çalışmada, çörek otu olarak bilinen Nigella sativa (N.sativa) tohumlarının kazlarda plazma malondialdehit (MDA), nitrik oksit (NO) ve tam kan glutatyon (GSH) düzeyleri üzerine etkilerinin araştırılması amaçlanmıştır. Bu amaçla, 8 adet Anser anser ırkı kaz 6 hafta boyunca % 5 öğütülmüş N. sativa tohumu içeren yemle beslendi. Çalışmanın başlangıcında ve her hafta başında olmak üzere 6 hafta boyunca kazların V. subcutenea ulnarisinden kan örnekleri usulüne uygun olarak alındı. Plazma MDA ve tam kan GSH düzeylerinin ölçümü sırasıyla Yoshioka ve ark ile Beutler ve ark’nın bildirdiği yönteme göre spektrofotometrik olarak yapıldı. Plazma NO düzeyleri ise Miranda ve ark’nın bildirdiği yönteme göre mikroplayt okuyucuda ölçüldü. Haftalara göre MDA, GSH ve NO düzeylerinde değişiklikler tespit edildi. Yapılan çalışmada elde edilen veriler 1. hafta ile kıyaslandığında N. sativa ilave edilen yemle beslenen kazlarda 2. haftadan itibaren plazma MDA düzeyle-rinin azaldığı, tam kan GSH düzeylerinin ise yükseldiği belirlenmiştir. Buna karşın haftalara göre plazma NO düzeylerinde bir fark tespit edilememiştir Sonuç olarak, kazlara 6 hafta süreyle uygulanan N. sativa’nın oksidatitif strese karşı koruyucu bir etkiye sahip olabileceği kanaatine varılmıştır.
