Determination of Neospora caninum and Toxoplasma gondii in Sheep and Goat Fetuses Using Dublex PCR, Immunohistochemistry, and Immunflourescence Methods

Abstract

Bacterial and viral agents have been commonly started to investigate in abortions which are seen in sheep and goats in our country. The study, which involves three different methods (dublex PCR, immunohistochemistry and immunofluorescence) onto sheep and goat abortions which are caused by Neospora caninum and Toxoplasma gondii agents has been never performed before. This study is aimed to determination of Neospora caninum and Toxoplasma gondii in sheep and goat abortions. Neospora caninum was not determined in samples of inspected 16 sheep and 7 goat abortions. However, Toxoplasma gondii was detected into one sheep abortion with dublex PCR, immunohistochemistry and immunofluorescence methods, it was also detected into one goat abortion with dublex PCR, immunohistochemistry methods. The antigens, which was immnunohistochemically positive detected in samples of sheep were observed into the liver, but they were observed into myocard in addition to liver in samples of goat abort. In contrast to any positiveness was not detected in dyeing that was performed with immunofluorescence methods, positiveness was observed into spleen of sheep abortions. Consequently, this project suggested that existence of Toxoplasma gondii has been first revealed by using duplex PCR together with immunohistochemistry and immunofluorescence methods in sheep and goat abortions in our country.

Özet

Ülkemizde koyun ve keçilerde görülen abortlarda genel olarak bakteriyel ve viral etkenlerin aranmasına gidilmiştir. Koyun ve keçi abortlarında Neospora caninum ve Toxoplasma gondii' nin etkenlerinin teşhisine yönelik dubleks PCR, immunohistokimya ve immunfloresans metotlarını içeren bir çalışma yapılmamıştır. Bu çalışmada koyun ve keçi abortlarında Neospora caninum ve Toxoplasma gondii varlığının tespit edilmesi amaçlanmıştır. İncelenen 16 koyun ve 7 keçi abort örneğinde Neospora caninum tespit edilmedi. T. gondii 1 koyun abortunda dubleks PCR, immunohistokimya ve immunfloresans yöntemleriyle, 1 keçi abortunda ise dubleks PCR ve immunohistokimya yöntemleriyle saptanmıştır. İmmunohistokimyasal olarak pozitif tespit edilen koyun abort örneğinde antijenler karaciğerde, keçi abort örneğinde ise karaciğere ek olarak, miyokardda görüldü. İmmunfloresans yöntemle yapılan boyamalarda keçi abortunda herhangi bir pozitiflik tespit edilmezken, koyun abortunda dalakta pozitiflik görüldü. Sonuç olarak sunulan çalışma ile T. gondii' nin koyun abortlarında Türkiye' de varlığı dubleks PCR, immunohistokimyasal ve immunfloresans yöntemleri birlikte kullanılarak ilk kez ortaya konulmuştur.
