Cytogenetic studies in patients with uremia before hemodialysis (A preliminary study)

Abstract
This study has been planned to elucidate the effect of uremia on human genome. For this purpouse, on the mitotic metaphases obtained from the cultured periferic blood samples of 18 patient with uremia to be given hemodialysis treatment for the first time and 15 healthy control subjects. Sister Chromatid Exchange (SCE) rates, numeric and structural chromosomal anomalies have been screened. SCE rate in patients with uremia was 9,48$pm$0,29 per metaphase where it was observed as 6,51$pm$0,24 per metaphases in healthy controls. Student-t test demonstrated that this difference was statistically significant (p=0.000). Standart cytogenetic analysis did not revealed any numeric or strucrural abnormalities in controls but in patient group some structural anomalies like chromosomal breaks, gaps, isogaps and acentric chromosomes were observed. These findings were supporting the idea that uremia may have some degrees of genotoxic effect.

Bu çalışma, üreminin genom üzerine olan etkilerini araştırmak için planlanmıştır. Bu amaçla, ilk kez hemodiyalize girecek üremili 18 hastadan ve 15 kontrol bireyinden alınan periferik kan örneklerinde "Sister Chromatid Exchange" (SCE) oranları ile sayısal ve/veya yapısal kromozom anomalileri sitogenetik teknikler kullanılarak araştırılmıştır. Üremili hastalarda metafaz başına SCE ortalaması 9,48±0,29 iken, kontrol grubunda bu sayı 6,51±0,24 olmuştur. Student-t testi kullanılarak yapılan istatistiksel değerlendirme sonucu her iki grup arasındaki farkın anlamlı olduğu tespit edilmiştir (p=0,000). Standart kromozom analizi çalışmalarında, kontrol grubundaki bireylerde herhangi bir kromozom anomalisi tespit edilememiştir. Üremili hastaların incelenen metafaz plaklarında; gap, izogap, asentrik kromozomlar ve kromozom kırığı şeklinde yapısal kromozom anomalileri tespit edilmiştir. Bu sonuçlar, üreminin genotoksik etkisi olabileceği düşüncesini desteklemektedir.
