Detection of antibodies against borrelia burgdorferi in cattle sera by fluorescent antibody technique for first time in Turkey

Abstract
The aim of this study, was to determine the presence of anti-B. burgdorferi antibodies by the use of indirect fluorescent antibody technique (IFAT) in cattle sera obtained from farms located in different regions. B. burgdorferi strain B31 was used for antigen preparation for IFA technique. The culture was diluted to give a final concentration of 50 organisms per 10 jul, and 30 /I of this suspension were placed on FA-slides, and kept frozen at -80°C until used. IFA technique was performed on 111 sera using a rabbit anti-bovine fluorescein isothiocyanate-labelled anti-IgG conjugate. Sera were tested at starting dilutions of 1:64 with subsequent two-fold dilutions up to 1:1024. Among 111 sera examined, 15 (13.5%) were found positive for antibodies to B. burgdorferi at a serum dilution of 1:256 or greater while, 96 (86.5%) sera were considered as negative. Among these 15 positive sera, 8 (53%), 3 (20%), and 4 (27 %) sampels were positive at serum dilutions of 1:256, 1:512, and 1:1024, respectively. According to these results, sero-positivity agonist B. Burgdorferi was detected in cattle sera examined.

Bu çalışmada, farklı bölgelerdeki çiftliklerden sağlanan 111 adet sığır kan serumunda B. Burgdorferi'ye karşı oluşan antikorlar indirekt flüoresan antikor tekniği (IFAT) ile saptanmıştır. B. burgdorferi B31 susundan antijen hazırlanarak kültür 50 mikroorganizma/10/id olacak şekilde sulandırılmış ve özel lamlara 30 jul miktarında damlatılıp, kullanılıncaya kadar -80°C'de saklanmıştır. Hastalıktan şüpheli sığırlara ait kan serumları 1/64'den 1/1024'e kadar iki katlı sulandırılmış ve fluorescein-isothiocyanate ile işaretli tavşan anti-sığır IgG konjugatı kullanılarak İFA tekniği ile test edilmiştir. Testte 1/256 ve üzerindeki serum türesi pozitif kabul edilmiştir. Test edilen serumlardan 15'i (%13.5) pozitif, 96'sı (%86.5) negatif bulunmuştur. Pozitif bulunan 15 'serumdan 8'i (%53) 1/256, 3'ü (%20) 1/512 ve 4'ü (%27) de 1/1024 titrede reaksiyon vermiştir. Bu sonuçlara göre, incelenen sığır serumlarında B. Burgdorferi yönünden seropozitifliğin bulunduğu saptanmıştır.
