Detection of antibodies of IBR-IPV infection in blood and milk sera by serum neutralisation test and virus isolation from milk samples dairy cows

Abstract
In this study, the presence of antibody against BHV1 in the samples of blood and milk obtained from 486 cows that 390 of'them were healty appearance or showed the symptoms of IBR-IPV infection and 96 were suffered from mastitis in 10 install type dairy farms located in various parts of Turkey were investigated by microneutralisation test. The usage of milk serum in the diagnosis of IBR-IPV infection as an alternative to blood serum was controlled by the microneutralisation test. The role of BHV1 in the samples of milk obtained from 96 cows with mastitis in epidemiology of the disease was examined. At the end of the experiment, 359 blood sera samples (74%) and 209 milk sera samples (43%) were found to be positive for BHV1 antibody. The number of cows whose both of the samples of milk and blood were positive for BHV1 antibody was 209. BHV1 was isolated from one of the milk samples obtained from 96 cows with mastitis. This is the first report about BHV1 isolation in milk sample from cow with mastitis in Turkey.
Özet
Bu araştırmada, Türkiye'nin değişik bölgelerinde kamu ve özel sektöre ait 10 adet kapalı süt sığırcılığı işletmesinde bulunan sağlıklı görünümlü ve IBR-IPV semptomları gösteren 390 adet ve mastitisli 96 adet olmak üzere toplam 486 adet inekten ayrı ayrı kan ve süt örneği alınarak, . mikronötralizasyon testiyle kan ve süt serumlarında BHV1 antikorlarının varlığı ve enfeksiyonun teşhisinde süt serumu örneklerinin kan serumu örneklerine alternatif materyal olarak kullanılabilirliği araştırılmıştır. Araştırmanın sonunda test edilen 486 adet kan serumu örneğinden 359 (%74) adedi, süt serumu örneğinin 209 (% 43) adedi BHV1 antikorları yönünden pozitif bulunmuştur. BHV1 antikorları yönünden her iki örneğin de pozitif bulunduğu hayvan sayısı 209 (% 43) olarak tespit edilmiştir. Bu araştırmada ayrıca süt örneklerinin IBR-IPV enfeksiyonunun epidemiyolojisindeki rolünün araştırılmasına yönelik olarak mastitisli ineklerden alınan 96 adet süt örneğinin bir adedinden virus izolasyonu gerçekleştirilmiş, SN ve EM sonuçlarına göre BHV1 olarak identifıye edilmiştir. Bu araştırma, Türkiye'de mastitisli ineklere ait sütten BHVl izolasyonunun gerçekleştirildiği ilk çalışmadır.
