Abstract

The diagnosis of visceral larva migrans in the experimentally infected mice with Toxocara canis was investigated comparatively by using indirect Haemagglutination (IHA) and indirect Flourescence Antibody (IFA) tests. The T. canis eggs used in the study were obtained from female parasites, which were collected from the intestines of dead dogs. Toxocara canis antigens used in the study were also obtained from adult parasites (female and male) which were collected from the intestines of dead dogs in the same way. Mice were infected by 2000 T. canis infected eggs. For every infection days 5 infected and 5 control (without T. canis infection) mice were used, 10 mice in all. After the 0., 2., 4., 6., 8., 10., 12., 14., 16., 18., 20., 22., 24., 26., 28., 30., 33., 36., 39., 42., 46., 50., 54., 58., 62., 75., 90. and 120th days of infection during the serological test by using both tests, first antigens were detected on zero day with I HAT and 14th day with I FAT after geometric average was taken. As a result of serological examinations, IF AT was found to be more specific but less sensitive than I HAT in the diagnosis of visceral larva migrans depending on T. canis. False positive reactions were observed both in IF AT and IHAT control group serums. However, in the IHAT, false positive reactions were found to be at higher levels than those in IF AT.

Bu çalışmada Toxocara canis ile deneysel enfekte farelerde indirekt hemaglutinasyon ve indirekt floresan antikor testleri ile karşılaştırmalı olarak visceral larva migransın teşhisi araştırılmıştır. Çalışmada kullanılan T. canis yumurtaları, ölü köpeklerin bağırsaklarından toplanan dişi parazitlerden, antijenler de hem dişi, hem de erkek T. canis'lerden elde edilmiştir. Visceral larva migransı oluşturmak amacı ile her fareye ortalama 2000 Toxocara canis yumurtası verilmiştir. Belirlenen her gün için 5 enfekte, 5 kontrol olmak üzere toplam 10 fare kullanılmıştır. Enfeksiyondan sonraki 0., 2., 4., 6., 8., 10., 12., 14., 16., 18., 20., 22., 24., 26., 28., 30., 33., 36., 39., 42., 46., 50., 54., 58., 62., 75., 90. ve 120. günlerde her iki testle de yapılan serolojik kontrollerde, geometrik ortalama çıkarıldıktan sonra, IFAT ile en erken 14. günde, I HAT ile ise 0. günde antikor tespit edilmiştir. Serolojik kontroller sonucunda, T. canis'den meydana gelen visceral larva migransın teşhisinde iFAT'ın daha spesifik, IH AT in ise daha duyarlı olduğu gözlenmiştir. Hem IFAT, hem de IHAT'da kontrol grubu serumlarda, yalancı pozitif reaksiyonlar gözlenmiştir. Ancak IHATdaki yalancı pozitif reaksiyonların I FAT'a göre çok daha yüksek oranda olduğu dikkat çekmiştir.
