Fertility obtained from frozen ram semen with different extenders containing varied antioxidants

Abstract

The aim of this research was to evaluate the fertility relying on in vitro and in vivo findings, after freezing ejaculates of Akkaraman rams with different extenders and antioxidants. In experiment, totaly 42 ejaculates collecting from rams per day by artificial vagina method were treated with regard to principle spermatological characteristics. Ejaculate volume averaged 1,04 ± 0,04ml, sperm motility 76.55±1.34 %, sperm concentration 2.60 ± 0.11 xl09/mi, the percentage of abnormal sperm 11.70 ±2.60 %, the percentage of dead sperm 9.63 ±0.93 % and sperm pH 6.75 ±0.05 were established in fresh semen.Sperm motility after thawing in frozen ram semen was averaged 48.30 ±2.13 % by the Tris-1 (vitamin C, glycerol, egg yolk), 54.33 ±1.88 % by the Tris-2 (taurine, glycerol, egg yolk), 43.33 ±1.88 % by the Tris-3( glycerol, egg yolk), 47.33 ±2.02 % by the Hepes-l( vitamin C, glycerol, egg yolk), 62.83 ±1.69 % by the Hepes-2( taurine,glyserol, egg yolk) and 44.33 ± 2.00 % by the Hepes-3( glycerol.The percentage of abnormal sperm after thawing was averaged 20.03 ± 1.23 %, 17.77 ± 1.20 %, 25.50 ± 7.55 %, 79.60 ± 1.13 %, J5.83 ± 1.39 % and21.83 ± 2.14 % for the same extenders respectively.Similary, the percentage of dead sperm was averaged 43.43 ±1.98 %, 34.36 ± 1.83 %, 46.77 ± 2.02 %, 44.83 ± 2.01 %, 32.77 ± 7.73 % and 49.30 ±2.14 % fa/-the same extenders respectively.In experiment, birth-rate was averaged 25.55 %, litter-size was averaged 1.2 and twinningrate was averaged 34.78 %.

Çalışmada Akkaraman ırkı koç ejakulatlannın farklı sulandırıcı ve antioksidanlarla dondurulmasından sonra, in vitro ve in vivo bulgulara dayanarak dölveriminin değerlendirilmesi amaçlanmıştır.Araştırmada, koçlardan s un'i vajen yöntemiyle hergün alınan toplam 42 ejakıdatta saptanan spermatolojik özelliklerden ejakulat miktarı 1,04 ±0,04 mi, spermatozoa motüitesi % 76,55 ±7,34, spermatozoa yoğunluğu 2.60 ±0.11 xl09/ml, anormal spermatozoa oranı % 11.70 ± 2.60, Ölü spermatozoa oranı % 9.63 ± 0.93 ve spermanın pH değeri 6.75 ± 0,05 olmuştur. Dondurulmuş koç spermalarında çözüm sonu genel ortalama spermatozoa motilitesi Tris-1 ile % 48.30 ± 2.13, Tris-2 ile % 54.33 ± 1.88, Tris-3 ile % 43.33 ± 1.88 olurken, Hepes-1 ile % 47.33 ± 2.02, Hepes-2 ile % 62.83 ± 1.69 ve Hepes-3 ile % 44.33 ±2.0 kaydedilmiştir. Çözüm sonu genel ortalama anormal spermatozoa oranı aynı sulandırıcılar için sırasıyla % 20.03 ± 1.23, %17.77 ± 1.20, % 25.50 ± 7.55, % 79.60 ± 1.13, % 15.83 ± 1.39 ve % 21.83 ± 2.14 saptanmıştır.Benzer şekilde çözüm sonu ölü spermatozoa oranı da sırasıyla % 43.43 ± 1.98, % 34.36 ± 1.83, % 46.77 ± 2.02, % 44.83 ± 2.01, % 32.77 ± 1.73 ve % 49.30 ± 2.14 bulunmuştur. Çalışmada genel ortalama doğum oranı %25.55, yavru oranı 1.2 ve ikizlik oranı ise %34.78 olmuştur.
