Investigation of biological control possibilities of wheat root rot disease caused by Bipolaris sorokiniana (Sacc.) using PGPR and bio-control bacteria in controlled condition

Abstract

The aim of this study was to determine the effect of using PGPR and biocontrol bacteria against root rot disease on wheat caused by Bipolaris sorokiniana (Sacc.) control and plant growth promotion. A total of 212 antagonistic bacterial isolates belonging to 25 different species isolated from under-ground or above-ground of wild and cultivated plants in the previous studies were used in this study, carried on in 2012-2013. They were identified using fatty acid methyl esters profiles by the Microbial Identification System (MIS). The antagonistic bacterial strains were tested to determine their antifungal properties against the fungal pathogen in vitro conditions. A total of 39 antagonistic bacterial isolates, selected by determining antifungal properties, nitrogen fixation and phosphate solubulising activities, were growth in suitable liquid carrier formulation. They tested for suppressing root rot disease and wheat plant growth parameters by using seed coating method in pot experiments. It was found that many of the antagonistik bacteria both suppressed the disease and provided important contributions to the development of the plant. Consequently our results indicated that some of the liquid formulation consist of antagonistic bacteria (Bacillus megaterium TV-6D, Brevibacillus choshinensis TV-53D and Bacillus pumilus TV-3C) can be used as both biopesticide for the control of wheat root rot disease and biofertilizers for wheat crop.

Özet

Bu çalışmanın amacı; PGPR ve biyoajan bakteriler kullanılarak buğday kök çürüklüğü hastalığı Bipolaris sorokiniana (Sacc.)’nın kontrolü ve bitki gelişimi üzerine etkisinin belirlenmesidir. 2012-2013 yıllarında yapılan bu çalışmada; daha önce yapılmış olan farklı araştırmalarda yabani ve kültür bitkilerinin toprak altı veya toprak üstü aksamlarından izole edilerek, Mikrobiyal Tanı Sistemi (MIS) ile tanılanan 25 faklı türe ait toplam 212 antagonist bakteri izolatı kullanılmıştır. Bu antagonist bakteriler in-vitro koşullarda patojene karşı antifungal özelliklerinin belirlenmesi için test edilmiştir. Antifungal özellikleri, azot fiksasyonu ve fosfatı çözebilme özellikleri dikkate alınarak seçilen 39 antagonist bakteri izolatı sıvı taşıyıcı formülasyonda geliştirilmiştir. Tohum kaplama yöntemi kullanılarak saksı denemelerinde hastalık gelişimi ve buğday bitkisinin gelişim parametreleri üzerine etkinlikleri bakımından test edilmiştir. Antagonistik bakterilerin pek çoğunun hem hastalığı baskılamakta etkili olduğu hem de bitki gelişiminde önemli katkılar sağladığı tespit edilmiştir.
