Method of measuring transient evoked otoacoustic emissions in Guinea pigs

Abstract
Objectives: To develop a reliable method of measuring transient evoked otoacoustic emissions (TEOAE) in guinea pigs and to compare guinea pig TEOAEs with those of human ears. Design and Methods: We measured TEOAEs in 15 normal guinea pig ears with ILO88 (ver 4.20B). Transient responses to non-linear clicks were analyzed during the first 20 milliseconds (ms) after stimulus onset. The time window was then set from the appearance time of TEOAEs to 10 ms, the original waveform was subsequently reconstructed. To ensure that actual biologic responses and TEOAEs were recorded, we compared (i) emissions obtained from live animals and from those which were rendered dead through overdoses of anesthetic drugs, (ii) and those obtained before and after exposure of a 1 kHz pure tone or wide-band noise of 110 dB for 10 minutes. To compare guinea pig TEOAEs with those of humans, emissions were recorded from 15 healthy adult ears. Results: Control comparisons between both live and dead animal emissions and recordings before and after noise exposure showed that TEOAE recordings were accurate in guinea pigs, with elimination of artifacts of adaptors used for better ear compliance and echo of the stimuli in the ear canal. Mean latencies of dominant emission and endpoint time of TEOAEs were 2.34±0.67 and 5.80±0.79 ms in guinea pig ears, and 6.53±1.52 and 17.58±1.35 ms in human ears, respectively. Conclusion: As guinea pig TEOAEs are shorter in duration than those of humans, and latencies of dominant emission are usually less than 2.5 ms, the reconstruction technique used in our study seems to be more reliable and practical to measure TEOAEs in the guinea pig compared with the original measurement between 2.5-20 ms.

Amaç: Kobaylarda transient evoked otoakustik emisyonlar (TEOAE) için bir ölçüm yöntemi belirlemek ve kobay ile insan TEOAE'lerini karşılaştırmak. Çalışma Planı ve Yöntemler: Normal işitme saptanan sekiz kobayın (guinea pig) 15 kulağında ILO88 (ver 4.20B) cihazı ile TEOAE'ler ölçüldü. Kullanılan yöntemde, ilk 20 milisaniyedeki (msn) orijinal ölçüm, emisyonun başlangıç süresi ile 10. msn arasında rekonstrükte edildi. Ölçümlerin gerçek biyolojik cevabı yansıttığı ve TEOAE'leri doğru olarak ölçtüğünü saptamak için, canlı ve anestezik ilaçla yaşamları sonlandırılan kobaylar arasındaki emisyonlar ve 10 dk süreyle 110 dB, 1 kHz saf ton veya 10 dk süreyle 110 dB wide-band akustik uyaran öncesi ve sonrası ölçümler karşılaştırıldı. Kobay ve insan TEOAE'lerinin karşılaştırılması için, aynı cihazla 15 normal işitmeli, erişkin insan kulağındaki emisyonlar ölçüldü. Bulgular: Canlı-ölü ve gürültü öncesi-sonrası ölçümler sonucu, probun kobay kulağına uyumu için kullanılan adaptörlerin artefaktının ve stimulusun kulak yolundaki ses yansımasının elimine edilerek, TEOAE'lerin doğru olarak kaydedildiği gösterildi. Dominant TEOAE latans ve emisyon bitiş süreleri ortalaması sırasıyla, kobaylarda 2.34±0.67 ve 5.80±0.79 msn, insanlarda 6.53±1.52 ve 17.58±1.35 msn bulundu. Sonuç: Kobaylarda TEOAE'lerin insanlara göre daha kısa süreli olması ve dominant emisyon latans ortalamasının 2.5 msn'nin altında olması nedeniyle, insanlarda kullanılan 2.5-20 msn arasındaki ölçüm yerine çalışmamızda kullanılan rekonstrüksiyon tekniğinin, kobay TEOAE ölçümünde daha gerçekçi bir yöntem olduğunu düşünüyoruz.
