The effects of xylometazoline and pseudoephedrine on rat nasal and tracheal mucosa

Abstract

Objectives: We investigated the effects of topical and systemic decongestant preparations on rat nasal and tracheal mucosa. Design: Twenty-six male Wistar rats were assigned to three groups. The first group (n=10) was given oral pseudoephedrine (80 microliters} twice daily for 15 days. The second group (n=10) was administered intranasal topical xylometazoline (10 microliters} twice daily for 10 days. All medications were given under ether anesthesia. Six rats in the control group were subject to only ether anesthesia once daily. At the end of the administration, sections from the anterior and posterior nasal mucosa and trachea were obtained and stained with hematoxylin and eosin. Histopathologically, intraepithelial gland proliferation, goblet cell proliferation, the severity of inflammatory cell infiltration, fibrosis, and mucosal thickness were evaluated. Results: No significant changes were observed between the two groups with regard to mucosal edema; however, mucosal thickness differed remarkably when compared with the controls, showing a significant decrease, and a significant increase, respectively, (p<0.05). In the second group, significant loss of cilia and increase in goblet cells were observed in sections from the posterior turbinates. Discussion: Both pseudoephedrine and xylometazoline seem to exert undesirable effects on the nasal and tracheal mucosa.

Özet

Amaç: Bu çalışmada topikal ve sistemik dekonjestan preparatların sıçan nazal ve trakeal mukozasındaki etkileri araştırıldı. Çalışma Planı: Wistar cinsi 26 erkek sıçan üç gruba ayrıldı. Birinci gruptaki 10 sıçana, 15 gün süreyle günde iki kez 80 mikrolitre dozda oral psödoefedrin, ikinci gruptaki 10 sıçana 10 gün süreyle günde iki kez 10 mikrolitre dozda intranazal topikal ksilometazolin eter anestezisi altında uygulandı. Üçüncü gruptaki altı sıçan ise kontrol grubu olarak ayrıldı. Kontrol grubuna günde bir kez sadece eter anestezisi yapıldı. Çalışma sonunda histopatolojik İnceleme için sıçanların ön ve arka nazal mukozaları ile trakealarından geçen kesitler hazırlandı, hematoksilen-eozin ile boyanarak incelendi. Kesitlerde İntraepitelyal gland artışı, goblet hücre proliferasyonu, enflamatuar hücre infiltrasyonunun şiddeti, fibrozis ve mukoza kalınlığı değerlendirildi. Bulgular: İki dekonjestan grubu arasında mukozal ödem açısından anlamlı farklılık bulunmadı. İkinci grupta arka/posterior konka kesitlerinde epitel hücrelerinde anlamlı derecede silia kaybı ve goblet hücre artışı görüldü. Her iki grup arasındaki mukoza kalınlığı da belirgin derecede farklı idi. Birinci grupta kontrol grubuna göre anlamlı derecede mukozal incelme, ikinci grupta kalınlaşma gözlendi (p<0.05). Tartışma: Psödoefedrinin ve ksilometazolinin nazal ve trakeal mukoza üzerinde istenmeyen etkileri olduğu sonucuna varıldı.
