The in vitro and in vivo effect of water of Lake Van on pH ,buffer capacity and total titrated acidity of rumen fluid in sheep

Abstract
The effect of Van Lake water on rumen fluid, pi I, total titratable acidity and buffer capacity in sheep was investigated in two phases.The study svas earned out on 162 rumen fluid samples obtained from 67 sheep. In in vitro phase of the study, the samples of rumen fluid were diluted in the ratio of 1/20,-1/10, 1/5 and 1/1 with Van Lake water After dilution, the rumen fluid pl 1 and buffer capacity have increased (p<0.05-0.001) and total tilratable acidity has decreased (p<0.05-0.001), significantly. Above dilution of Van Lake water had increased rumen fluid pll (p<0.001 ) significantly which was reduced to 5.5. 5.0 and 4.5 by lactic acid. In m vivo phase of the study; Van Lake water (one liter lor each) was given to healthy sheep with rumen lube. Thereafter, the rumen fluid pH and buffer capacity had increased (p<0.001j and total titratable acidity had decreased (p<0.001) significantly m the first 30 minutes. Rumen fluid pll and buffer capacity had decreased (p<0.001), total Utralable acidity had increased (p<0.001) significantly in the first 2 hours for the 1 group (received saccharose at the level of 10 g/kg bods weight in 1 liter water) and 2nd group (received saccharose at the level of 1 lg/kg body weight in 1 liter ssater), and in the 6-12th hours for the 3 group (received crushed svheat at the level of 40 g/kg body weight). The rumen fluid pH and buffer capacity had increased (p<0.01-0.001 ı and total titratable acidity had decreased (p<0.01-0.001) significantly in the first hours after Van Lake water (3 liters for each sheep) was given to all groups. After 3 liters Van Lake water administration orally for 3 group, increase in pH and buffer capacity (p<0.01-0.001) and decrease in total titratable acidity (p<0.01-0.001) during 1st hour have remained unchanged trough 6th and 12th hours while those parameters returned normal values ssithm 24 hour. Based on above results, it has convinced that the therapeutic and prophylactic effect of Van Lake Water on ruminal acıdosis should need more comprehensive investigations

Özet: Van Gölü Suyunun koyunlann rumen sıvısı pH, total asidite ve tampon kapasitesi düzeylerine etkisi iki aşamada araştırıldı. Çalışma, 67 koyundan alman 162 rumen sıvısı örneği üzerinde yürütüldü. Araptyrmanm in vitro adamasında, Van Gölü Suyu ile 1/20, 1/10, 1/5 ve 1/1 oranlarında sulandırılan rumen sıvısı örneklerinde, pH ve tampon kapasitesinin artığı (p< 0.05-0.001), total asidite miktarının azaldığı ( p<0.05, p<0.001) saptandı. Van Gölü Suyu; laktik asit ile pITsı 5.5, 5.0 ve 4,5'e indirgenen rumen sıvısı örneklerinde de aynı oranlarda sulandırmadan sonra, pH artışı (p<0.001) sağladı. İn vivo aşamada; birer litre Van Gölü suyu içirilen sağlıklı koyunlann ilk yarım saatte rumen sıvısı pH ve tampon kapasitesinin arttığı (p<0.001), total asidite miktarının azaldığı )p<0.01) belirlendi. Rumen sıvısı pll, total asidite ve tampon kapasitesi normal sınırlarda bulunan 1. grup (10 gr/kg şeker eriyiği), 2. (inip (11 gr/kg şeker enyiği) ve 3. Grup (40 gr/kg kınlmış buğday verilen) koyunlarda ilk 2 saatte (1 Ve 2. Grup) ve 6-12. Saatte (3. Grup), pH ve tampon kapasitesinin düştüğü (p<0.001). total asidite miktarının yükseldiği (P<0.001) saptandı. Her 3 grup koyunda 3'er litre Van Gölü suyu içirildikten sonraki 1. Saate pH ile tampon kapasitesinin yükseldiği (p<0.01-0.001), total asıdite miktarının düştüğü (p<0.01-0.001), 6. ve 12. Saatlerde de bu durumun devam ettiği ve 24. Saatte parametrelerin normal sınırlarda olduğu belirlendi. Sonuç olarak; rumen sıvısı pil ve tampon kapasitesinde artışa total asidite miktarında ise azalmaya neden olan Van Gölü suyunun, rumen asiditesinin sağaltım ve profilaksisindeki etkisinin daha kapsamlı bir şekilde araştırılmasının yararlı olabileceği kanısına varıldı.
