The effects of cyclosporine A, ascorbic acid and alpha tocopherol on vasodilatation responses due to acetylcholine (ACh) in rabbit thoracic aorta

Abstract

In the present in-vitro study, the effects of Cyclosporin A (Cs A), ascorbic acid (vitamin C) and a-tocopherol (vitamin E) on the acetylcholine (Ach)-induced relaxation responses in the rabbit thoracic aorta were investigated. Also vitamin C and Vitamin E effects, on cyclosporin A- induced effects had been histo-pathological differences in the rabbit liver and kidneys were investigated. In this study there were four groups: control group (n=6), applied Cs A (n=6), applied Cs A+VitC+VitE (n=8) and applied VitE+VitC (n=6). During ten days, drugs were applied in all groups except the control group. In all experimental groups maximum contractile responses, that were ma¬intained by contracting thoracic aortas with 3x10~5 M noradrenaline(NA) were taken as 1009c Re¬laxation responses in all groups, which were cu¬mulative applied of Ach (10~9- 10'4 M), were found as 94 %, 279c, 649o control group, Cs A + vit C + vit E, vit C + vit E groups respectively. .Acetylcholine did not cause relaxation response in the applied CsA group. In the presence of L-NAME (NG-nitro-L-arginine metlyester) (10^ M), cont¬ractile responses due to NA was significantly inc¬reased in all groups except applied CsA group (p<0.05). Also inhibition responses due to Ach was inhibited in all groups by L-NAME (10~4 M). CsA induced some histo-pathological differences on the rabbit liver and kidneys. Vitamin C and Vi¬tamin E were partly prevented from these changes. In the rabbit thoracic aorta, cyclosporin A comp¬letely interrupted the relaxation responses to Ach and Vit C and Vit E, which have antioxidant effect, partly prevented this effect.

Özet

Sunulan bu invitro çalışmada tavşan torasik aor¬tasında asetilkolin (Ach) ile oluşan gevşeme ce¬vapları üzerine Siklosporin A (Cs A), askorbik asit (Vit.C) ve a-tokoferol Vit.E'nin etkileri araştırılmıştır. Ayrıca siklosporin A'nın tavşan ka¬raciğer ve böbreklerinde oluşturduğu histopaıolojik değişiklikler üzerine vitamin C ve vitamin E' nin et¬kilerine bakılmıştır. Çalışma Cs A uygulanan (n=6), Cs A+ vitamin C+ vitamin E uygulanan (n=8); vitamin E+ vitamin C uygulanan (n=6) ve kontrol grubu (n=6) olmak üzere 4 grupta yapılmıştır. Kontrol grubunun dışındaki diğer gruplara 10 gün süre ile ilaç uy¬gulanmıştır. Tüm deney gruplarında torasik aortalar 3xlO~5 M noradrenalin (NA) ile kasılarak elde edi¬len maksimum kasılına cevapları 9c 100 kabul edil¬miş ve kümülatif uygulanan ACh (10~9- 10-4 ^j j]e kontrol grubunda %94' I ilk, Cs A+ vitamin C+ vi¬tamin E verilen grupta %27' lik ve vitamin E+ vi¬tamin C verilen grupta 9c64' Kik bir gevşeme cevabı alınmıştır. Cs A verilen grupta ise ACh' ne gevşeme cevabı alınmamıştır. NA ile elde edilen kasılma ce¬vapları ortama N&- nitro- L- arginin metil ester (L-NAME, 10"* M) ilavesi ile CsA verilen grup dışında diğer gruplarda anlamlı olarak artmıştır (p<0.05). Ayrıca 10~4 M L-NAME, ACh ile elde edilen gevşeme cevaplarını tüm gruplarda inhihe etmiştir. CsA karaciğer ve böbrek dokusunda his-topatolojik değişikliklere neden olmuş, vitamin C ve vitamin E ise bu değişikliklere kısmen engel olmuştur. Elde edilen sonuçlar, CsA tavşan, torasik aortasında ACh bağlı gevşeme cevaplarını tamamen ortadan kaldırırken, antioksidan etkili olan askorbik asit ve a-tokoferol buna tamamen olmasa da kısmen engel olmaktadır.
