The effects of ethanol on development and morphogenesis of the mammalian embryos at the stage of early organogenesis, and the role of free radicals in this effect

Abstract

Aim: The excessive maternal alcohol drinking results in Fetal Alcohol Syndrome which is a pathology caused by the embryotoxic effects of ethanol in prenatal period. The mechanisms of ethanol action at the cellular or molecular levels are scarce. In this study it was aimed to investigate the effects of ethanol on growth and development of mammalian embryos as well as the morphological structure, and the role of free radicals on these effects. Methods: Wistar rats were obtained from the Selçuk University Experimental Medicine Research Center, and their 9.5 days embryos were explanted and cultured for 48 hours in rat serum. Whole rat serum was used as a culture medium for the control group while different concentrations of ethanol (250-500 mg%) were added to rat serum for the experimental groups. Also, the lowest effective concentration of ethanol for all parameters was added to the culture media in the presence of an antioxidant superoxide dismutase (SOD). Ten embryos were used for each experimental condition. Dose-dependent effects of ethanol on embryonic developmental parameters such as; total morphological score, yolk sac diameter, crown-rump length, somite number, embryo and yolk sac protein contents were compared using morphological and biochemical methods. Each embryo was evaluated for the presence of any malformations. Results: Compared to the control embryos, ethanol significantly decreased all developmental parameters dosedependently (p<0.05) with an increase in overall dismorphology. The haematoma in different regions, especially in the neural tube was most frequently observed. When the SOD was added to the culture media in the presence of ethanol, growth and developmental parameters were improved (p<0.05) and there was a decrease in the incidence of malformations (p<0.05). Conclusion: A dose dependant developmental toxicity of ethanol on rat embryos during organogenesis was determined, these effects might involve free oxygen radicals.

Özet

Amaç: Gebe kadınların aşırı alkol almaları Fötal Alkol Sendromu adı verilen ve etanolun prenatal dönemde embriyotoksik etkileri sonucu ortaya çıkan bir patolojiye neden olmaktadır. Etanol'ün bu dönemde hücresel ve moleküler düzeydeki etki mekanizmaları ise tam olarak bilinmemektedir. Bu çalışmada etanolun embriyonik büyüme ve gelişme ile morfolojik yapı üzerine olan etkileri ve serbest radikallerin bu etkilerdeki rolünün araştırılması amaçlandı. Yöntem: Selçuk Üniversitesi Deneysel Tıp Araştırma ve Uygulama Merkezi hayvan laboratuvarından elde edilen Wistar ratlardan gebeliklerinin 9.5 uncu gününde diseke edilerek çıkartılan embriyoların rat serumu içerisinde 48 saat süreyle kültürü yapıldı. Kültür ortamı olarak; kontrol grubu için normal rat serumu kullanılırken, deney grupları için rat serumuna değişen konsantrasyonlarda etanol (250-500 mg%) ilave edildi. Ayrıca etanol'ün toksik etkisinin tüm parametreleri etkilediği dozu ile birlikte antioksidan superoksid dismutaz (SOD) kültür ortamına ilave edildi. Her bir konsantrasyon için l0 rat embriyosu kullanıldı. Etanol'ün rat embriyolarının gelişim parametreleri (total morfolojik skor, yolk saik çapı, tepe-kıç mesafesi, somit sayısı, embriyo ve yolk saik protein içerikleri) üzerine doz bağımlı etkileri, morfolojik ve biyokimyasal yöntemlerle karşılaştırıldı. Embriyolar malformasyon varlığı acısından da değerlendirildi. Bulgular: Kontrol embriyoları ile karşılaştırıldığında etanol'ün doz bağımlı olarak bütün gelişimsel parametreleri gerilettiği (P<0.05] ve genel morfolojide bozukluklara sebep olduğu gözlendi. Özellik/e nöral tüp olmak üzere değişik bölgelerde hematomlar gözlendi. Kültür ortamına etanol ile birlikte SOD eklendiğinde büyüme ve gelişme parametrelerinde artış (P<0.05j ve malformasyon sıklığında azalma (P<0.05) tespit edildi. Sonuç: Etanol'ün organogenez dönemindeki rat embriyoları üzerine doz bağımlı gelişimsel toksisiteye sebep olduğu ve bu etkilerde serbest oksijen radikallerinin rol oynayabileceği belirlendi.
