A research on the anatomical and histological investigations of bud union at T-Budding in budded chestnut

Abstract
This study was carried out to determine the anatomical and histological properties of bud union at some chestnut types obtained by selection between 1996-1998. With this purpose, seeds taken from totally 21 chestnut types from seven provinces belonging to Aegean Region were used as production material. The seedlings belonging to the types that were proper for budding were budded by T-Budding Technique and the cross-sections were taken to investigate the bud union as anatomically and histologically at three different times, such as, 30, 60 and 90 days after budding. At cross-sections taken after 30 days from budding, bud union between rootstock and scion were occured by two seperate callus tissue originated from two budding parts, and at cross-sections taken after 60 and 90 days after budding, it was observed that vascular connection was provided by the development of bud union.
Özet
Bu çalışma ile bazı kestane tiplerinde aşı kaynaşmasının anatomik ve histolojik özelliklerinin belirlenmesi amaçlanmıştır. 1996-1998 yılları arasında yürütülen bu denemede Ege Bölgesi'ne ait yedi ilden toplam 21 adet kestane tipine ait tohumlar materyal olarak kullanılmıştır. Bu tiplerin tohumlarından yetiştirilen çöğürlerden aşılama büyüklüğüne ulaşanlar, T-göz aşısı yöntemi ile aşılanmışlar ve aşı kaynaşmasının anatomik ve histolojik gelişimini incelemek amacıyla 30, 60 ve 90 gün sonra enine kesitler alınmıştır. Aşılamadan 30 gün sonra alınan enine kesitlerde; bütün tiplerde birleşmenin her iki aşı elemanından ayrı ayrı oluşan kallus dokusu aracılığı ile gerçekleştiği; 60 ve 90 günlük kesitlerde, bu kaynaşmanın gelişerek vasküler irtibatın kurulduğu görülmüştür.
