Expression of hepatitis B "e" antigen gene in Escherichia coli

Abstract

In this study, the "e" antigen gene region of hepatitis B virus (HBV) was cloned and expressed to Escherichia coli in order to prepare the antigens used in HBeAg and anti-HBe tests for the serodiagnosis of HBV infection. Firstly, the primers coding the "e" proteinic region of HBV were detected and the "e" genomic region of HBV were amplified by PCR from HBV-DNA. Then the recovered amplified product was cloned to pCR-TOPO plasmid via the TOPO-TA (Invitrogen) cloning kit. The HBe gene region cut by EcoRI and Sail was cloned to the pSE420 (4.6kb-lnvitrogen) expression plasmid. Finally, this plasmid was transferred to Ecoli and the expressed HBeAg was detected and confirmed by enzyme immunoassay (ELISA).

Özet

Bu çalışmada hepatit B virus (HBV) enfeksiyonunun tanısında kullanılan serolojik yöntemlerden HBeAg ve anti-HBe için gerekli antijenin hazırlanması amacıyla HBV'nin "e" antijen gen bölgesi Eschericha coli'ye klonlanmış ve eksprese edilmiştir. Bunun için öncelikle HBe protein bölgesini kodlayan genin primerleri tesbit edilmiş ve HBV DNA'sından PCR metoduyla HBe gen bölgesinin PCR ürünü elde edilmiştir. Elde edilen amplifikasyon ürünü TOPO TA (Invitrogen) klonlama kiti ile pCR-TOPO plazmidine klonlanmıştır. Buradan EcoRI ve Şali ile kesilen parça pSE420 (4.6kb-lnvitrogen) ekspresyon plazmidine klonlanmıştır. E.coli'ye transfer edilen plazmid ile HBeAg'nin ekspresyonu sağlanmıştır. Daha sonra HBeAg varlığı enzim immünoassay (ELISA) ile teyit edilmiştir.
