Cloning of Hepatitis B virus core antigen (HBcAg) gene in Escherichia coli

Abstract
In this study, hepatitis B virus core antigen (HBcAg) gene was cloned in E. coli by using recombinant DNA technology. In the first stage of the study, hepatitis B virus (HBV) DNA's have been isolated from the sera of HBV-DNA positive patients, and 550 base pair gene region which encodes HBcAg, was amplified by polymerase chain reaction (PCR). Later, construction of recombinant plasmid containing HBcAg gene, transformation into E.coli, and identification of recombinant plasmids have been performed. The plasmid vector (pUC 19) and HBcAg gene were digested by EcoRI and Hind III restriction endonucleases. HBcAg gene was cloned into plasmid vector directionally and the plasmid pHBC-1 contanining HBcAg sequences was used in the transformation of E.coli. The transformed bacterial cells were selected onto ampicillin containing media. Presence of HBcAg gene in the recombinant plasmid (pHBC-1) was confirmed by PCR and restriction endonuclease analysis.

Bu çalışmada, Hepatit B virüsünün "core" antijen (HBcAg) geni rekombinant DNA teknolojisi kullanılarak Escherichia coli'de klonlanmıştır. Bu amaçla çalışmanın ilk aşamasında, hepatit B virüsü (HBV) ile enfekte bireylerin serum örneklerinden HBV DNA'sı izole edilmiş ve bu DNA'dan HBcAg'ni kodlayan, 550 baz çifti uzunluğunda gen bölgesi polimeraz zincir reaksiyonu (PCR) ile çoğaltılmıştır. Daha sonra, HBcAg genini içeren rekombinant plazmidin oluşturulması, E.coli'ye transformasyonu ve rekombinant plazmidlerin tanımlanması yapılmıştır. Plazmid vektör (pUC 19) ve HBcAg geni EcoRI ve Hind III restriksiyon endonükleaz enzimleriyle sindirilmiş, HBcAg geni plazmide yerleştirilmiş ve oluşturulan pHBC-1 plazmidi E.coli hücrelerinin transformasyonunda kullanılmıştır. Transformasyona uğrayan E.coli hücreleri ampisilin içeren ortamda seçilmiş ve rekombinant plazmidlerin (pHBC-1) HBcAg genine sahip oldukları PCR ve restriksiyon enzim analizi yapılarak doğrulanmıştır.
