Isolation and identification of staphylococci from raw milk samples and investigation of slime factor in coagulase-negative strains

Abstract

Staphylococci which are widely distributed in nature, human and animal flora, are the most important causative agents of nosocomial and community acquired infections. As they possess a variety of pathogenicity factors, the spectrum of staphylococcal infections is quite large and infections transmitted by food have a special value. The aim of this study was to isolate and identify staphylococcus species from raw milk samples, and to search for the production negative staphylococci. Among the 52 raw milk samples collected between October 1999-March 2000 period, 38 (73%) of them yielded staphylococci and 16 (42.1%) of these were coagulase positive, while 22 (57.9%) were coagulase negative. Coagulase positive staphylococci were identified as S.aureus (n: 11) and S.intermedius (n: 5). Among the coagulase negative staphylococci identified, 2 strains of S.simulans, 1 strain of S.lugdunensis, 1 strain of S.delphini and 1 strain of S.auriculahs were found to produce slime factor. As a result, consumption of unpasteurized milk or milk products contaminated with coagulase positive and/or negative staphylococci, from animals with mastitis or from normal flora of handlers of dairy cows, would be a potential infection source, although not frequently encountered as the other pathogens transmitted via milk.

Doğada, insan ve hayvanların florasında yaygın olarak bulunan stafilokoklar, nozokomiyal ve toplumdan kazanılmış enfeksiyonların en önemli etkenlerindendir. Çok sayıda patojenite faktörüne sahip olan stafilokokların enfeksiyon spektrumu oldukça geniş olup, bunlar arasında besinlerle bulaşan enfeksiyonlar önemli yer tutmaktadır. Bu çalışmada, önemli besin maddelerinden biri olan çiğ süt örneklerinden stafilokok türlerinin izolasyonu ve tanımlanması ile koagülaz negatif stafilokoklarda (KNS) bir virülans faktörü olarak kabul edilen slime maddesi üretiminin araştırılması amaçlanmıştır. Ekim 1999-Mart 2000 tarihleri arasında 52 çiğ süt örneği çalışılmış ve bunların 38 adedinden (%73) stafilokok türleri izole edilmiştir. İzole edilen suşların 16'sı (%42.1) koagülaz pozitif (KPS), 22'si (%57.9) KNS olarak tanımlanmış ve KPS'larm 111 S.aureus, 5'i S.intermedius olarak tiplendirilmiştir. KNS'lardan 5'inde (%22.7) ise (2 S.simulans, 1 S.lugdunensis, 1 S.delphini, 1 S.auricularis) slime faktör üretimi saptanmıştır. Sonuç olarak gerek süt veren hayvanlarda mastit etkeni olan gerekse süt üretimi ile uğraşan kişilerin florasında bulunan koagülaz pozitif ve/veya negatif stafilokok türleri ile kontamine olmuş ve pastörize edilmemiş ürünlerin tüketilmesi, sütle bulaşan diğer patojenler kadar sıklıkla olmasa da potansiyel bir enfeksiyon kaynağı oluşturacaktır düşüncesindeyiz.
