Isolation of mesophilic Aeromonas species from mussels and determination of cytotonic enterotoxin gene by polymerase chain reaction

Abstract
This study was carried out to isolate Aeromonas spp. from 515 mussel samples collected from Keban Dam Lake and to determine the presence of cytotonic enterotoxin gene, a virulence factor detected in A. hydrophila, in the isolates by polymerase chain reaction (PCR). Homogenized soft tissues of mussel samples were inoculated onto blood agar media and biochemical tests were performed on the suspected colonies. Aeromonas spp were isolated from 3.5% (18/515) of the samples, and 12 (2.3%) were identified as A. hydrophila, 3 (0.6%) were identified as A. cavia and 3 (0.6%) were identified as A. sobria. The DNA samples were extracted from these isolates and amplified by PCR using a pair of primers derived from the cytotonic enterotoxin gene of A. hydrophila. In the agarose gel examination of the PCR products, 2.7% (14/ 515) of the isolates were determined to have this virulence factor. It was concluded that the quantitative data provided in this study was of significance for public health due to the fact that mesophilic Aeromonads can cause serious disorders, such as gastroenteritis, wound infections and septicaemia in humans.

Bu çalışmada, Keban Baraj gölünün iki farklı bölgesinden toplanan 515 midye örneğinden Aeromonas türlerinin izolasyonu ve izolatlarda virulans faktörlerinden sitotonik enterotoksin geninin polimeraz zincirleme reaksiyonu (PZR) ile saptanması amaçlanmıştır. Midyelerin yumuşak dokularından hazırlanan homojenizatlar kanlı agara ekilmiş ve üreyen şüpheli kolonilere biyokimyasal testler uygulanmıştır. Örneklerin %3.5'inden (18/515) Aeromonas türleri izole edilmiş, bunların 12'sinin (%2.3) A.hydrophila, 3'ünün (%0.6) A.cavia ve 3'ünün (%0.6) de A.sobria olduğu belirlenmiştir. İzolatlardan ekstrakte edilen DNA'lar A.hydrophila'da tespit edilen sitotonik enterotoksin geninden türetilen bir çift primer kullanılarak PZR'de amplifikasyona tabi tutulmuş ve PZR ürünlerinin agaroz jel elektroforezde değerlendirilmesi sonunda %2.7'sinde (14/515) bu virulans geninin mevcut olduğu saptanmıştır. Çalışmada izole edilen mezofilik Aeromonas etkenlerinin, insanlarda gastroenteritler başta olmak üzere yara enfeksiyonları ve septisemi gibi hastalıklara neden olduğu dikkate alındığında, elde edilen kantitatif verilerin insan sağlığı açısından önem taşıyacağı düşünülmüştür.
