Changes in blood, brain and hippocampal lipid peroxidation and antioxidant enzyme activity in cigarette smoke exposed rats

Abstract
In this study we investigated the effect of cigarette smoke on lipid peroxidation and antioxidant enzyme activity in the blood, brain and hippocampus of rats. Rats were divided into two groups. These groups are : Group 1, comprised of only diet feeding, was served as control and Group 2, comprised of diet feeding and exposed to cigarette smoke, was served as test group. Rats were exsplosed to cigarette smoke for 2 hours/day during 4 weeks. At the end of the experiment, malondialdehyde (MDA) and glutathione (GSH) concentrations, and antioxidant enzyme activities were assayed in erythrocyte, brain, and hippocampus samples. Statistically significant difference in erythrocyte, MDA levels between the two groups was found (p<0.05). Erythrocyte SOD and catalase activities were significantly lower in the experiment group thanin the control group (p<0.05). As a result, we thought that cigarette smoke caused an oxidative strees in the body, but the brain and hippocampus were not influenced from cigarette smoke.
Özet
Sigara dumanına maruz bırakılan ratlarda lipid peroksidasyonu ve antioksidan enzim aktiviteleri araştırıldı. Bu maksatla Wistar Albino türü ratlarda 2 grup oluşturuldu. 1. Grup sadece yemle beslenen ratlardan oluşup kontrol grubu olarak kullanıldı. 2. grup yemekle birlikte sigara dumanına maruz bırakıldı ve deney grubu olarak kullanıldı. Deney grubundaki ratlar 4 hafta süresince günde toplam 2 saat sigara dumanına maruz bırakıldı. 4 hafta sonunda kan, beyin ve hipokampüs örnekleri alınarak, antioksidan enzim aktiviteleri, glutatyon (GSH) ve malondialdehit (MDA) düzeyi ölçüldü. Eritrosit MDA düzeyi kontrol grubuna göre deney grubunda kontrol grubuna göre anlamlı şekilde düşük bulundu (p<0.05). Eritrosit SOD ve katalaz aktiviteleri ise deney grubunda kontrol grubuna göre anlamlı şekilde düşük bulundu (p<0.05). İki grup arasında beyin ve hipokampüs MDA ve GSH konsantrasyonları ile antioksidan enzim aktivitelerinde fark görülmedi. Sonuç olarak sigaranın vücutta oksidatif stres oluşturduğu, ancak beynin sigaranın oluşturduğu oksidatif stresten etkilenmediği düşünüldü.
