Effects of N-acetylcysteine and L-carnitine on erythrocyte antioxidant systems in experimental colitis

Abstract

Ulcerative colitis is an inflammatory bowel disease. In recent years, It has been suggest that free oxygen radicals play the role in the pathogenesis of inflamed mucosa. The aim of the present study was to investigate antioxidant effects of N-acetylcysteine (NAC) and L-carnitine (LCAR) on the erythrocyte antioxidant systems of rats in the acetic acid-induced experimental colitis model. Materials and Methods: Colitis was induced in rats by intrarectal administration of 1 ml of 4% acetic acid (AA) in the experimental groups. NAC an LCAR treatment, 500 mg/kg was administrated in a 1 ml volume by intraperitoneal route. Rats were divided into four group; Group 1: Intrarectal and intraperitoneal saline were administrated as a control group, Group 2: Intrarectal AA was administrated as a colitis model, Group 3; Intrarectal AA administration 1 hour after the intraperitoneal administration of NAC as a pretreated-NAC group, Group 4; intrarectal AA administration 1 hour after the intraperitoneal LCAR administration of as a pretreated-LCAR group. Rats were sacrificed after 24 hours following the NAC and LCAR treatment and then the blood were taken. The levels of glucose-6-phosphate-dehydrogenase (G6PD), catalase (CAT), superoxide dismutase (SOD), glutathione-S-transferase (GST) and also glutathione (GSH) such as antioxidant systems were measured in the erythrocytes. Results: While G6PD enzyme activity increased in colitis group, The activities of CAT, SOD, GST, and the levels of GSH decreased compared to control group (p<0.05). In the treatment groups, the activities of SOD and GST, and the level of GSH were found as high compared to the colitis group (p<0.05). While CAT activity increased in LCAR group (p<0.05), It didn't change in the NAC group (p>0.05). However, while G6PD activity decreased in NAC group, It didn't change in LCAR group. Conclusions: Oxidative stress decreased in the colitis for which NAC and LCAR changed the balance of the living antioxidant systems. For this reason, NAC and LCAR could be beneficial as a complementary agent in ulcerative colitis treatment.

Özet

Ülseratif kolit, inflamatuvar bir barsak hastalığıdır. Son zamanlarda mukozanın inflamasyonunun patogenezinde serbest oksijen radikalleri üzerinde durulmaktadır. Bu çalışma, ratlarda asetik asit (AA) ile oluşturulan deneysel kolit modelinde N-asetilsistein (NAC) ve L-karaitin (LCAR)'in eritrosit antioksidan sistemler üzerine etkilerini araştırmak amacıyla yapılmıştır. Gereç ve Yöntem: Çalışma gruplarında kolit modeli 1 mi %4'lük AA intrarektal yoldan verilmesi ile oluşturulmuştur. NAC ve LCAR tedavisi, 500 mg/kg intraperitoneal 1 ml olarak verilmiştir. 10'ar rattan oluşan 4 grup şu şekilde belirlenmiştir. Grup 1: İntrarektal serum fizyolojik verilen kontrol grubu, Grup 2: İntrarektal AA verilen kolit grubu, Grup 3: Kolit oluşturulmadan 1 saat önce NAC verilen tedavi grubu ve Grup 4: Kolit oluşturulmadan 1 saat önce LCAR verilen tedavi grubu. Ratlar, akut kolit oluşturulduktan sonra ve/veya tedavi verildikten 24 saat sonra sakrifıye edilerek kanları alındı. Eritrositlerde antioksidan sistemlerden; glukoz-6-fosfat dehidrogenaz (G6PD), katalaz (CAT), süperoksit dismutaz (SOD), glutatyon S-transferaz (GST) aktiviteleri ve glutatyon (GSH) düzeyleri ölçüldü. Bulgular: Kolit grubunda kontrol grubuna göre G6PD enzim aktivitesi artarken CAT, SOD ve GST aktiviteleri ile GSH düzeylerinin anlamlı olarak düştüğü gözlenmiştir (p 0,05). Bunun yamsıra, NAC grubunda G6PD aktivitesi kolit grubuna göre düşük iken (p 0,05). Sonuç: NAC ve LCAR canlı antioksidan sistemlerinin dengesini değiştirerek kolitin oluşturduğu oksidatif stresi azaltmaktadır. Bu nedenle NAC ve LCAR, ülseratif kolitte yardımcı bir tedavi seçeneği olabilir.
