Investigation of reduced glutathione (GSH), malondialdehyde (MDA) and nitric oxide (NO) levels in ewes receiving zing during the period of pregnancy

Abstract

This study was aimed to investigate the effect of zinc on reduced glutathione (GSH), malondialdehyde (MDA) and nitric oxide (NO) levels of ewes during pregnancy. Twenty Tuj breed ewes at 3 years of age were used as animal material. The animals were divided into 2 equal groups including control (n=10) and experimental group receiving 30 mg/kg of zinc daily for average of 150 days (n=10). The blood samples from the groups were monthly collected before (once) and during the pregnancy (for five months). The whole blood GSH, plasma MDA and NO levels were spectrophotometrically determined. Ewes in both groups had higher MDA and decreased GSH levels during the pregnancy as compared to the period before pregnancy (p<0.001). As of the second month of pregnancy, ewes receiving zinc had lower MDA but higher GSH concentrations for the 4 consecutive months compared to control (p<0.01) until the end of experiment (5th month). Nitric oxide level of both groups wh,ich was raised with the beginning of pregnancy reached to its peak level on the third month of pregnancy. In the zinc-received group, the NO levels were found to be lower than that of control group on the fifth month of pregnancy (p<0.05). In conclusion, oral administration of zinc to pregnant ewes could improve altered antioxidant status and contribute to the reduced levels of NO elevated during the last stage of pregnancy.

Özet

Çalışmada gebelik periyodu boyunca koyunlara çinko verilmesinin, redükte glutatyon (GSH), malondialdehit (MDA) ve nitrik oksit (NO) düzeyleri üzerine etkisinin araştırılması amaçlandı. Çalışmada materyal olarak yaklaşık 3 yaşlı 20 adet Tuj ırkı koyun kullanıldı. Koyunlar deneme (n=10) ve kontrol (n=10) olmak üzere iki eşit gruba ayrıldı. Deneme grubundaki koyunlara gebelik periyodu süresince (ortalama 150 gün) her gün 30 mg çinko ağız yolu ile verildi. Gebe kalmadan önce bir kez ve gebelik süresince her ay kan numuneleri alındı. Numunelerde tam kan GSH düzeyleri ile plazma MDA ve NO derişimleri spektrofotometrik olarak ölçüldü. Gebelik öncesine göre gebelik boyunca her iki grupta da MDA düzeyleri artarken, GSH düzeyleri azaldı (p<0.001). Gebeliğin ikinci ayından itibaren çinko verilen grupta kontrole göre MDA düzeyleri azalırken, GSH düzeyleri artış gösterdi (p<0.01). Gebelikle birlikte artmaya başlayan NO değerinin gebeliğin 3. ayında en yüksek düzeyde olduğu saptandı. Çinko verilen grupta ise NO düzeylerinin gebeliğin 5. ayında kontrol grubuna göre düşük olduğu tespit edildi (p<0.05). Sonuç olarak; gebe koyunlara ağız yolu ile verilen çinkonun gebelikte değişen antioksidan sisteminin güçlenmesine ve gebeliğin son döneminde artan NO düzeyinin azaltılmasına olumlu katkılar sağladığı sonucuna varıldı.
