Producing Longlasting Education Material For Oncology By Prolonged Tissue Processing: Preliminary Study

Abstract

Objective: The aim of this research is, to produce organs and tissues as a nontoxic, dry, odorless, touchable education material for pathology and oncology departments of medical faculty and oncology institutes. German Professor von Hagen invented plastination. This new method enables to preserve tissue material for longer time. In this method, the water inside the tissues is replaced with special polymers and the final form of tissues resembles to its original form and can be preserved for educational purposes and biology museums. This method is expensive and requires special equipment. The staff who is performing the plastination is exposed to toxic substances for long periods. Tissue processing is used in pathology departments every day in routine practice. Tissue samples, less than 3 mm in thickness, are placed in cassettes. Then they are step-by-step left in formalin, increasing alcohol series, acetone, xylene and lastly in paraffin for 6 to 18 hours, dehydrated, hardened.and embedded in paraffin blocks to be sectioned for histologic evaluations. Method: In this study, biologic materials that are suitable for different education material cut from pieces with colon carcinoma, larynx carcinoma, breast carcinoma and renal carcinoma that were discarded one year after termination of routine processes were exposed to prolonged closed tissue processing machine with vacuum system. ranging from 3 to 10 days depending on the size of the samples, and different experimentation times. Results: The samples saturated with paraffin were prepared as hard, odorless, easily handled dried solid education materials. There were no putrefaction, shrinkage or deformation of the samples during their waiting periods ranging from 3 to 6 months. The samples were used as a preliminary trial in Basic Oncology master and doctorate lectures. Conclusion: Our future plans include experimenting with larger-sized and also rarely seen tumor samples, preparation education material for experimental animal studies,; embedding in transparent material such as epon, glass, durability tests, and testing the possibility of performing molecular experiments with stored samples. In addition, the durability of the material will be observed for a longer period.

Özet

Amaç: Bu çalışmanın amacı, Onkoloji Enstitüsü ve Tıp Fakültelerinin Patoloji ve Onkoloji bölümleri için, dokuları ve organları kuru, kokusuz, dokunulabilir ve toksik olmayan eğitim materyali olarak hazırlamaktır. Plastinasyon, Alman Profesör Von Hagen tarafından keşfedilmiş, doku örneklerinin uzun süreli koruyan bir yöntemdir. Bu teknik ile dokular arası su, özel polimerler ile yer değiştirerek, doğal hallerine bire bir benzer, örnekler eğitim amaçlı ve biyoloji müzeleri için elde edilmektedir. Bu yöntem pahalı ve özel ekipmanlar gerektiren, işlemi yapan teknisyenin uzun süre toksik maddelere maruz kalabildiği bir yöntemdir. Doku takibi rutin histopatoloji pratiğinde her gün kullanılan bir yöntemdir. Kasetlenmiş 3 mm kalınlığı geçmeyen doku örnekleri toplam 6-18 saat gibi bir sürede formol, artan alkol serileri, aseton, ksilol ve parafin aşamalarında belirli süreler kalarak doku mumyalanmakta ve parafin bloklara kesit alınmak üzere gömülmektedir. Yöntem: Bu araştırmada, eğitim materyali olabilecek bir kolon kanseri, bir larinks kanseri, bir böbrek kanseri, bir meme kanseri, bir yumuşak doku sarkomundan farklı parçalar rutin işlemler tamamlandıktan bir yıl sonra atılmak için ayrılan örneklerden boyuta ve farklı deneme sürelerine göre 3-10 gün süreli, otomatik vakumlu kapalı sistem doku takip cihazında uzatılmış doku takibine maruz bırakılmıştır. Bulgular: Parafine doymuş olarak çıkan örnekler kokusuz elle rahat ellenebilen kuru eğitim materyali olarak hazırlanmıştır. Örneklerin 3-6 aylık bekleme sürelerinde bozulma, kuruma deformasyon göstermediği saptanmıştır. Örnekler temel onkoloji yüksek lisans ve doktora derslerinde ön deneme olarak kullanılmıştır. Sonuç: Gelecek planlarımızda daha büyük örneklerle çalışma, nadir tümörleri çalışma, deney hayvanları için eğitim materyali hazırlama, cam ve reçine gibi materyallere gömme ve dayanıklılık testleri ve saklanan örneklerden moleküler çalışma olasılığının test edilmesi vardır. Ek olarak, materyal dayanıklılığı daha uzun süre gözlenecektir.
