The Application Fields in Parasitology of RFLP Technique

Abstract

Restriction endonucleases which cut the short double-stranded DNA sequence at their specific regions and remove the target genes from these regions are multifunctional enzymes. These enzymes also known as molecular scissors are examined including four general groups such as type I, type II, type III and homing endonuclease. Tip-II endonucleases that fully capable of cutting from the target zone are the most preferred enzymes in especially cloning studies. RFLP technique is the process for separation of different-sized fragment DNA molecule by using restriction enzymes. This technique is a popular method widely used for obtaining recombinant DNA, the creation of restriction map, identification of population polymorphism, the diagnosis of genetic diseases. In recent studies, this technique often has been preferred for diagnosis of protozoa and differentiation of species due to simple, fast and reliable a technique.

Özet

Restriksiyon endonükleazlar, çift iplikli kısa DNA dizilerini özgül bölgelerden keserek hedef gen bölgesinin çıkartılmasını sağlayan multifonksiyonel enzimlerdir. Moleküler makaslar olarak da bilinen bu enzimler tip-I, tip-II, tip-III ve homing endonükleazlar olmak üzere dört genel grupta incelenir. Bugün kullanılan enzimlerin çoğu tam hedef bölgeden kesim yapabilen ve özellikle klonlama çalışmalarında tercih edilen tip-II endonükleazlardır. RFLP tekniği ise restriksiyon enzimleri kullanılarak DNA molekülünün farklı büyüklükteki fragmentlere ayrılması ve görüntülenmesi işlemidir. Rekombinat DNA elde edilmesinde, restriksiyon haritalarının oluşturulmasında, popülasyon polimorfizmlerinin belirlenmesinde ve genetik hastalıkların teşhisinde yaygın olarak kullanılan popüler bir yöntemdir. Son zamanlarda yapılan çalışmalarda, protozoonların tanısında ve tür ayrımında da basit, hızlı ve güvenilir bir teknik olması nedeniyle sıklıkla tercih edilmeye başlanmıştır.
