Abstract

OBJECTIVE: The aim of our study is to make wild type p53 gene transfection to the human epithelial ovarian cells by using strontium phosphate, lacking the potential disadvantages of liposome and viral vectors as toxicity and genetical changes. Thus, an experimental model is constructed to study the effect of gene transfection, and synergistic effect of the other treatment modalities on cell proliferation. MATERIAL AND METHOD: The p53 gene mutant human ovarian carcinoma cell line (MDAH 2774) was obtained from American Type Culture Collection. Cells were thawed and cultured in RPMI-1640 that contains 10 % Fetal Bovine Serum (FBS). They were incubated with strontium phosphate integrated wild type p53 DNA at 37 °C and under 5%CO<SUB>2</SUB> pressure for 16-24 h. On the third day of the transfection cells were washed with PBS twice and fixed by 0.2% gluteraldehide. To determine the efficacy of transfection &beta;-galactosidase activity was evaluated. Cells incubated with strontium phosphate only were used as negative control. Immunofleuroscence was done to show the protein expression of transfected wild type p53 DNA. RESULTS: Cultured human ovarian epithelial cells were successfully transfected with strontium phosphate in vitro, p-galactosidase activity confirmed the successful transfection and protein expression of the transfected gene was confirmed by immunofleuroscence. There was no signal in the cells incubated with strontium phosphate only. CONCLUSION: In this study we have transfected the wild type p53 DNA to MD2774 human ovarian cancer cell line by strontium phosphate lacking the genetic disadvantages of viral vectors and toxic effects of the liposomes. Moreover, this model may have some advantages not only to study the consequences of gene transfection, but also synergistic effects of the medical treatments on ovarian cancer growth.

Çalışmamızın amacı insan epitelyal över kanseri hücrelerine stronsiyum fosfat kullanarak gen nakli yapmaktır. Bu yöntemle lipozomların toksik etkisinden ve viral vektörlerin neden olduğu genetik değişikliklerden kaçınılarak salt transfeksiyonun etkisini saptamak açısından bir model oluşturulmuştur. Bu model üzerinde hücre proliferasyonu üzerinde etkili olabilen diğer tedavi yöntemlerinin sinerjistik etkilerini daha, doğru bir şekilde araştırma olanağı da elde edilebilecektir. p53 geni mutant olduğu önceden bilinen insan över kanseri serisi (MDAH 2774) American Type Culture Collection'dan (ATCC) elde edilmiştir. Dondurulmuş hücreler çözülmüş ve %10 fetal bovine serum (FBS) içeren RPMI-1640. içerisinde kültürleri yapılmıştır. Stronsiyum fosfata bağlanmış sağlam (wild) p53 DNA'sı ile 37°C'de %5 CO 2 basınç altında 16-24 saat inkübe edilmişlerdir. Transfeksiyonun 3.günü hücreler phosphate buffered saline (PBS) ile 2 defa yıkanmışlar ve %0.2'lik gluteraldehit ile fikse edilmişlerdir. Transfeksiyonun etkinliği β-galaktosidaz aktivitesi ile araştırılmıştır. Sadece stronsiyum fosfat ile inkübe edilen hücreler negatif kontrol olarak kullanılmıştır. Transfekte edilen sağlam p53 DNA'sının protein ekspresyonu ise immunofleuroscence yapılarak araştırılmıştır. Kültüre edilmiş insan epitelyal över hücreleri stronsiyum fosfat ile, in vitro olarak başarıyla transfekte edilmişlerdir. Saptanan β-galaktosidaz aktivitesi stronsiyum fosfat ile yapılan transfeksiyonun başarısını kanıtlamıştır. Transfekte edilen p53 DNA'sının protein düzeyindeki sentezi ile immunofteuroscence ile gösterilmiştir. Sadece stronsiyum fosfat ile inkübe edilen hücrelerde ise sinyal saptanmamıştır. Çalışmamızda sağlam p53 DNA'sı literatürde ilk kez stronsiyum fosfat kullanılarak MD2774 insan över kanseri hücrelerine başarıyla transfekte edilebileceği gösterilmiştir. Bu yöntemle lipozomların toksik etkisinden ve viral vektörlerin neden olduğu genetik değişikliklerden kaçınılarak salt yapılan transfeksiyonun etkisini araştırabilme olanağı doğmuştur. Buna ek olarak, hücre proliferasyonu üzerinde etkili olabilen diğer tedavi yöntemlerinin etkinliğini daha doğru bir şekilde araştırma olanağının da elde edilebileceğini düşünmekteyiz.
