Abstract
OBJECTIVE: To investigate the biologic action of parathyroid hormone related protein (PTH-rP) in oocyte maturation. STUDY DESIGN: To evaluate if this peptide has a role on oocyte maturation, we used Xenopus laevis oocyte model. In this model criteria for oocyte maturation was germinal vesicle break down which can be observed as a white spot on the black pole of the oocyte. RESULTS: We found very high levels of parathyroid related protein (PTH-rP) in human and bovine follicular fluid. We showed on xenopus model that there is an inhibitory action of PTH-rP that is present in the follicular fluid to progesterone induced maturation of oocytes. CONCLUSION: PTH-rP secreted from the granulosa cells may be the oocyte maturation inhibitor (OMI) or one of them. There is need for further studies on the subject.
Özet
Biz çalışmamızda insan ve. sığır follikül sıvısında yüksek seviyelerde paratiroid hormon benzer peptit seviyeleri bulduk. Bu yüksek peptit seviyelerinin oosit matürasyonunda rolü olup olmadığını değerlendirmek için Xenoupus Leavis oosit modelini kullandık. Bu modelde oosit matürasyon kriteri olarak oositin siyah yüzünde beyaz leke şeklinde beliren germinal vesikül break down kullanıldı. Sonuç olarak PTH-rP progesteronun oluşturduğu oosit matürasyonunu doza bağlı inhibe etmektedir. Bzim düşüncemize göre granülosa hücrelerinden salınan bu peptit oosit matürasyon inhibitörü (OMI) veya bunlardan biridir.
