Comparison of the levels of endothelin-1 in aqueous humor pseudoexfoliation glaucoma, pseudoexfoliation syndrome, and normal subjects

Abstract

Purpose: A prospective, comparative study was carried out to investigate the concentration of endothelin-1 (ET-1) in the aqueous humor (HA) of cases patients with pseudoexfoliation glaucoma (PXG) and, pseudoexfoliation syndrome (PEX) and in normal subjects, and to compare the results with each other. Materials and Methods: HA samples were obtained from 13 eyes with PXG, 18 eyes with PXS, and 14 eyes of normal subjects at the beginning of trabeculectomy, phakoemulsificationphacoemulsification, or combined surgery. ET-1 concentration in HA was measured by enzyme-linked immunoassay. Mann-Whitney U test and Kruskal Wallis analysis of variance test were used for the statistical analysis. Results: The mean HA concentration of ET-1 was 7.49±1.05 pg/ml in the eyes with PXG, 7.45±1.41 pg/ml in the eyes with PXS, and 3.92±1.83 pg/ml in the control eyes. The ET-1 levels of both PXG and PXS cases patients were found to be statistically significantly higher than those of the control subjects (p<0.01); but no significant difference was found between PXG and PXS cases patients (p=0.95). Conclusion: Our results support a role for high ET-1 HA concentration in the etiopathology of PXG and PXS. However, no difference between the HA levels of ET-1 of the eyes with PXG and PXS was detected and further investigations with a larger number of cases were planned.

Amaç: Psödoeksfoliatif glokom (PEG), psödoeksfoliatif sendrom (PES) ve normal olgularda, aköz humor endotelin-1 (ET-1) düzeylerinin kıyaslanması. Gereç ve Yöntem: PEG’lu 13 göz, PES’lu 18 göz ve 14 normal gözden, trabekülektomi, fakoemulsifikasyon-intraoküler lens implantasyonu veya kombine glokom ve katarakt cerrahileri esnasında, aköz humor örnekleri alındı. ET-1 konsantrasyonları enzim-linked immünoassay metodu ile ölçüldü. İstatistiksel analizlerde, Mann-Whitney U testi ve Kruskal Wallis varyans analizi kullanıldı. Bulgular: Ortalama humor aköz ET-1 düzeyi PEG’lu gözlerde 7.49±1.05 pg/ml, PES’lu gözlerde 7.45±1.41 pg/ml ve normal gözlerde ise 3.92±1.83 pg/ml olarak saptandı. PEG ve PES’lu gözlerdeki humor aköz ET-1 düzeyleri arasındaki fark istatistiksel olarak anlamlı bulunmazken (p=0.95), her iki gruptaki değerlerin kontrol grubundaki gözlerin humor aköz ET-1 düzeylerine oranla istatistiksel olarak anlamlı ölçüde yüksek olduğu görüldü (p<0.01). Sonuç: Yüksek aköz humor ET-1 düzeylerinin, PEG ve PES patogenezinde rol oynayabileceği sonucuna gidildi. Ancak, PEG ve PES’lu gözler arasında anlamlı fark saptanamaması üzerine, gelecekte daha çok sayıda olguyu içeren çalışmaların yapılması planlandı.
