Serum hemocysteine, vitamin B6, B12 and folic acid levels in a rat model of intestinal ischemia-reperfusion

Abstract

Aim: Glutathione increases during ischemia-reperfusion (IIR) and shows many effects in the pathogenesis of this process. One of its effects is to modulate cysteine, which has an important role in the production of homocysteine in the cells. In this experimental study, the levels of homocy¬steine during intestinal IIR and the vitamins that play a role in its metabolism were investigated. Material and Method: Forty male Wistar-albino prepubertal rats weighing between 90 and 120 gr. were divided into 4 groups. Group 1 (control, n=10) was taken as control. Blood samples were taken without any applied procedure. In group 2 (sham, n=10) abdomen was opened, su¬perior mesenteric artery (SMA) was dissected and then closed. In Group 3 (ischemia, n=10) SMA blood flow was stopped for three hours. In Group 4 (IIR, n=10) the blood flow was maintained again after cessation for 3 hours and blood samples were taken after 3 hours of reperfusion. In all samples, plasma homocysteine, vitamin B6, B^ and folic acid levels were determined. The results were assessed with One-way ANOVA, Scheffe and Tukey HSD tests. Results: The increase of homocysteine levels was significant in ischemia and IIR groups when compared to control and sham groups (p<0.05). However the comparison of re¬sults between the ischemia and IIR groups was not significant (p>0.05). The difference of the vitamin levels was not significant between the groups, either (p>0.05). Conclusion: Homocysteine levels increase during ischemia. The levels of vitamins which take part in homocysteine metabolism do not change in the early period. The cutoff levels obtained during intestinal ischemic events can be used to determine the depth of ischemia in clinical practice.

Özet

Amaç: İskemi/reperfüzyon (IIR) sürecinde glutatyon miktarı artar ve birçok etki gösterir. Bu etkilerden birini de hücrelerde homosistein oluşumunda yer alan sisteini etkileyerek gösterir. Bu deneysel çalışmada intestinal UR sırasında, homosistein düzeyleri ve metabolize edilmesinde rol oynayan vitaminlerin durumu araştırıldı. Gereç ve Yöntem: Ağırlıkları 90-120 gr. arasında değişen, toplam 40 erkek Wistar-albino cinsi prepubertal rat 4 gruba ayrıldı. Grup 1 (kontrol, n=10) kontrol olarak alındı. Bunlara herhangi bir işlem uygulanmadan kan örnekleri alındı. Grup 2 (sham, n=10)'de karın açıldı, superior mezenterik arter (SMA) disseke edilip karın kapatıldı. Grup 3 fiskemi, n=10) SMA kan akımı üç saat süre ile durduruldu. Grup 4 (UR, n=10) üç saat durdurulan SMA kan akımı tekrar başlatıldı ve 3 saatlik reperfüzyonun ardından kan örnekleri alındı. Tüm örneklerde plazma homosistein, vitamin B6 ve Bi2, folik asit düzeylerine bakıldı. Sonuçlar tek yönlü AN OVA, Scheffe ve Tukey HSD testleriyle değerlendirildi. Bulgular: İskemi ve UR gruplarında homosistein düzeyleri kontrol ve sham gruplarından anlamlı oranda yüksek bulundu (p 0.05). Vitamin düzeyleri arasındaki farklılık da istatistiksel olarak anlamsız bulundu (p>0.05). Sonuç: Homosistein düzeyi iskemi periyodunda artmaktadır. Homosisteinin metabolizmasında rol oynayan vitaminlerin düzeyleri erken dönemde değişmemektedir. İntestinal iskemik süreçlerde elde edilecek eşik değerler ile klinikte iskeminin derinliğinin belirlenmesi mümkün olabilir.
