Abstract

Several lines of evidence support the notion that developmental process may be important in the etiology of autistic disorder. The aim of this paper is to review the neurodevelopmental process and the reported neuropathological findings in autistic disorder that may be a consequence of disturbances in this process. Method: Selected papers and books regarding to autistic disorder, neuronal migration, synaptic pruning and apopitosis are reviewed, and the consequences are summarized in presented review. Results: Many regions of the central nervous system have been implicated in the genesis of autism, but the neurobilogical basis of the disorder remains unknown. Investigation of central nervous system in autism gives impression of the disorder may be a neuronal circuitry disorder rather than involvement a specific brain region. Neuropathological, neurobiological, biochemical, sleep, structural and functional neuroimaging studies suggested that abnormal neuronal migration, synaptic pruning and apopitosis may play a role in the etiology of autistic disorder. Conclusion: The research to date has been hindered by methological difficulties and the heterogeneity of autism. Longitudinal studies in autism are needed to confirm a potential defect in neuronal migration, synaptic pruning and apopitosis during early developmental of life.

Özet

Amaç: Çeşitli kanıtlar otistik bozukluğun etiyolojisinde gelişimsel sürecin önemli olabileceğini desteklemektedir. Bu yazının amacı, nörogelişimsel süreç ve otistik bozuklukta bu sürecin sonucunda oluşabilecek nöropatolojik bulguları gözden geçirmektir. Yöntem: Bu derlemede, otistik bozukluk, nöronal göç, sinaptik budanma ve programlı hücre ölümüyle ilgili seçilmiş yazı ve kitaplar gözden geçirildi ve sonuçlan özetlendi. Sonuç: Otizmin meydana gelmesinde bir çok merkezi sinir sistemi bölgesi düşünülmüşse de, bozukluğun nörobiyolojik temeli bilinmemektedir. Otizmde, merkezi sinir sistemi araştırmaları, bozukluğun özgül bir beyin bölgesi tutulumundan çok, nöronal bağlantı bozukluğu olduğu izlenimini vermektedir. Nöropatoloji, nörobiyoloji, biyokimya, uyku, yapısal ve fonksiyonel nörogörüntüleme çalışmaları, anormal nöronal göçün, sinaptik budanmanın ve programlı hücre ölümünün otistik bozukluğun etyolojisinde rol oynayabileceğini düşündürmektedir. Tartışma: Günümüze kadar yapılan çalışmalarda yöntemsel güçlükler ve otizmin heterojenitesi nedeniyle sorunlar vardır. Otistik bozuklukta, yaşamın erken dönemlerindeki nöronal göç, sinaptik budanma ve programlı hücre ölümünde olası bozukluğu doğrulayacak izlem çalışmalarına ihtiyaç vardır.
