Abstract

Background and Aim: The aim of this study was to evaluate expression of receptor activator of nuclear factor kappa-B ligand (RANKL) and osteoprotegerin (OPG) in rat periodontal ligament (PDL) cells in response to mechanical force both in vivo and in vitro. Materials and Methods: In the in vivo study, a traction force of approximately 200 g was applied in the buccopalatal direction using an elastic rubber band. In the in vitro study, PDL cells were obtained from other rats’ (n=5) incisors and cultured 3-dimensionally in type-1 collagen gel. A closed glass receptacle containing water weighing a total of 25 g was placed on the collagen gel culture to apply approximately 5 g/cm2 of pressure for 24 hr. Results: In the in vivo experiment, at 7 days, PDL cells were positive for RANKL and OPG in the central area of the PDL and close to cementum. However, OPG positive cells gradually increased, whereas RANKL decreased. After the loss of the elastic rubber band’s elasticity, OPG positive cells began to increase due to initiation of regeneration and subsequent recovery of PDL cells. In the in vitro study, mRNA expression of RANKL showed small amount and decreased with time and there was no significant difference at any of the time period comparing to the control group. Expression of OPG, however, showed an increase after 1 day due to cellular response to pressure. Conclusion: These results suggest that RANKL and OPG are regulators of homeostasis of PDL and its surrounding tissues during orthodontic tooth movement.

Özet

Amaç: Bu çalışmanın amacı, mekanik kuvvete cevap olarak rat periyodontal ligament (PDL) hücrelerinde Hücre Reseptör Aktivatör Faktörü Kappa-B Ligand (RANKL) ve Osteoprotegerin (OPG) salınımının değerlendirilmesidir. Gereç ve Yöntem: In vivo çalışmada, bukko-palatinal yönde 200 g’lık traksiyon kuvveti uygulandı. In vitro çalışmada, diğer ratların (n=5) kesici dişlerinden PDL hücreleri alındı ve Tip-1 kolajen jel içerisinde 3-boyutlu olarak kültür edildi. Yaklaşık 5 g/cm2 basınçla mekanik kuvvet uygulamayı taklit etmek için, 25 g ağırlığındaki kapalı cam bir kap kolajen jel kültürün üzerine 24 saat süreyle yerleştirildi. Bulgular: In vivo deneyde, 7. günde, PDL’nin orta bölgesinde ve semente yakın olan bölgede RANKL ve OPG pozitifti. Bununla birlikte, OPG pozitif hücreler kademeli olarak artarken, RANKL azaldı. Elastik bandın elastisitesi kaybolduktan sonra, PDL hücrelerinin iyileşmesine ve rejenerasyonun başlamasına bağlı olarak OPG pozitif hücreler artmaya başladı. In vitro çalışmada, mRNA’nın RANKL salınımının düşük olduğu ve zamanla azaldığı ve kontrol grubuna kıyasla hiçbir değerlendirme periyodunda anlamlı bir farklılık olmadığı görüldü. Bununla birlikte, basınca karşı hücre cevap oluşması nedeniyle 1 gün sonra OPG salınımı arttı. Sonuç: Bu bulgular, ortodontik diş hareketi sırasında RANKL ve OPG’nin, PDL ve çevre dokular için hemostaz regülatörleri olduğunu ileri sürmektedir.
