[Antimicrobial potentials of lactic acid bacteria ısolated from a nigerian menstruating woman]

Abstract
AIM: Racial differences affect the composition of lactic acid bacteria (LAB) in women’s vagina. However, the bacteria present in women’s vagina exert protective effect against invading uropathogens through production of several inhibitory compounds. The LAB composition of the vagina of a menstruating Nigerian woman was examined to detect any difference between the subject’s vaginal LAB flora and reported cases of women from western world and to investigate the antimicrobial activities of these lactic acid bacteria against potential uropathogens and enteropathogens with analysis of possible compounds that may be responsible for inhibition. METHODS: Informed consent was obtained from the subject. LAB were identified by partially sequencing the 16S rRNA gene. The organic acids were detected through High Performance Liquid Chromatography (HPLC) while the volatile compounds were detected by gas chromatography. The hydrogen peroxide production was assayed through enzymatic reactions. RESULTS: Enterococcus faecalis FAA025 and Streptococcus equines FAA026 were the only bacterial strains isolated. The two LAB strains inhibited the growth of all tested uropathogens and enteropathogens to remarkable degree. Both strains produced high quantities of lactic acid while high quantities of hydrogen peroxide, acetic acid and ethanol were only observed in Streptococcus equines FAA026. CONCLUSİONS: The results of this study suggest that in spite of absence of lactobacilli during menstruation in the subject, other LAB present (Enterococcus faecalis FAA025 and Streptococcus equines FAA026) can exert protective effects against invading uropathogens. Also, the LAB composition of the Nigerian woman is similar to her counterparts in the West.

AMAÇ: Irksal farklılıklar kadınların vajinasındaki laktik asit bakterilerinin (LAB) kompozisyonunu etkiler. Kadınların vajinasında bulunan bakteriler bazı baskılayıcı ürünler vasıtasıyla üropatojenlerin yayılmasına karşı koruyucu etki gösterirler. Adet gören Nijeryalı bir kadının vajinasının LAB kompozisyonu, bu kadının vajinal LAB florası ve bildirilmiş Batılı kadın vakalar arasında fark olup olmadığını ortaya koymak için ve bu LAB’lerin potansiyel üropatojenlere ve enteropatojenlere karşı aktivitelerini araştırmak için baskılamadan sorumlu olma potansiyeli olan muhtemel bileşikler analiz edilerek değerlendirildi. YÖNTEM: Denekten bilgilendirilmiş onam alındı. LAB 16S rRNA geninin parsiyel sekanslaması ile tanımlandı. Uçucu bileşikler gaz kormatografisi ile taranırken organik asitler “High Performance Liquid Chromatography (HPLC)” ile tarandı. Hidrojen peroksit üretimi enzimatik reaksiyonlarla test edildi. BULGULAR: Enterococcus faecalis FAA025 ve Streptococcus equines FAA026 izole edilen bakteri türleriydi. İki LAB türü, test edilen tüm üropatojenlerin ve enteropatojenlerin büyümesini dikkate değer düzeyde baskıladılar. Her iki tür yüksek miktarda laktik asit üretirken sadece Streptococcus equines FAA026’da yüksek miktarda hidrojen peroksit, asetik asit ve etanol gözlendi. SONUÇ: Bu çalışma; denekte adet görme sırasında laktobasillerin olmamasına rağmen varolan diğer LAB’lerin (Enterococcus faecalis FAA025 ve Streptococcus equines FAA026) üropatojenlerin yayılmasına karşı koruyucu etki gösterdiğini akla getirmektedir. Ayrıca, Nijeryalı kadının LAB kompoziyonu Batılı kadınlarla benzerdi.
