Abstract

OBJECTIVE: The objective of present study was to evaluate the effect of administering preoperative pregabalin, pregabalin and dexketoprofen, or placebo on haemodynamic response, postoperative analgesia, intraoperative opioid consumption, and patient satisfaction. METHODS: Ninety ASA I-II patients, aged 18-65 years, undergoing laparoscopic cholecystectomy were included in the study. Patients received either oral pregabalin 150mg (group P), pregabalin 150mg and dexketoprofen 25mg (group PD), or placebo (group C) 60 min before surgery. Anesthesia was induced with propofol, fentanyl, and vecuronium and maintained with propofol, remifentanyl, air and O2. HR, MAP, oxygen saturation, and use of intra-operative remifentanyl was recorded during the operation. Postoperatively HR, MAP, respiration rate, sedation scala, and VAS were assessed at 1, 2, 6, 12, and 24 hours rest. Patients satisfaction score, side effects, and total analgesic requirement in the first 24 hours after surgery were analyzed and compared between the groups. RESULTS: Patients in the PD group had significantly lower postoperative VAS scores, particularly at 1-2 hours. Total intra-operative opioid consumption in the PD group was significantly less than group P and C. Patient's satisfaction in PD group was increased in comparison to the other groups. CONCLUSION: We concluded that preemptive use of pregabalin combined with dexketoprofen is effective in suppressing haemodynamic response to intubation and providing intraoperative and postoperative analgesia in patients undergoing laparoscopic cholecystectomy.
