Abstract

The prevalance of type 2 diabetes mellitus (T2DM) is rapidly increasing in all over the world. Insulin resistance plays a major role in the pathogenesis of T2DM and the majority of patients with T2DM have insulin resistance. Several studies have shown that overexpresion of membrane glycoprotein plasma cell antigen 1 (PC-1) [ectonucleotide pyrophosphatase phosphodiesterase 1] in muscle, adipose tissue, fibroblasts, and other tissues associated with insulin resistance, obesity and T2DM. Moreover, either experimants in transgenic mice in vivo or in cultured cells in vitro, PC-1 overexpression impairs insulin stimulation of insulin receptor (IR) activation and downstream signaling. In addition, variants in PC-1 gene are associated with insulin resistance, T2DM, and obesity in various ethnic populations. Therefore, PC1 is a strong candidate protein that may play a role in the pathogenesis of T2DM. In this article, it was reviewed the role of PC-1 in insulin resistance.

Bununla birlikte, gerek in vivo transgenik farelerde gerekse in vitro hücre kültüründe aşırı düzeyde eksprese edilen PC-1, insülin reseptör aktivitesinde ve sinyal mekanizmasında bozulmaya yol açmaktadır. Bu makalede insülin direncinde PC-1'in rolü gözden geçirilmiştir.
