Abstract

The present study aimed to compare different canal sealers in different setting times in terms of the cytotoxicity they induce on 3T3 mouse fibroblasts. AH Plus, AH 26, Endomethasone N, RSA and Sealapex were put into glass moulds in 2 mm diameter and 10 mm length prepared previously after mixing under sterilized condition according to manufacturer's instructions. Test samples were divided into three groups; in the first group canal sealers were freshly prepared and in the others, canal sealers were set for 3-days and 7-days. Samples with different setting times were diluted in 10 ml culture medium for extraction periods of 1, 4, and 7 days. Extracts were taken after each dilution period, sterilized by 0.22 jjm filter, diluted 1:1, 1:2, 1:4, 1:8, and applied to 3T3 mouse fibroblasts during one-day incubation. MTS (tetrazolium salt) analysis was performed to evaluate viable cell rates. Different variables included in our study were statistically evaluated both in relation to each other and independently in terms of the cytotoxicity they induced. The results confirmed that all the sealer samples and each setting and extraction times, and each dilution showed statistically significant differences in relation to each other (p AH 26 > Endomethasone N > AH Plus > RSA according to their cytotoxicity levels. Furthermore, it was observed that as setting time and dilution rate increase, cytotoxicity decreases, while the longer the extraction time, the greater the cytotoxicity.
