Abstract

Soil salinity causes yield loss in plant production, but also directly or indirectly affects seed yield and quality. Iğdır plain has a high salinization potential due to hot and dry summers, low annual average rainfall for Turkey, and high groundwater. This situation has already caused some lands of the plain to be out of agriculture. In this study, it was tried to determine some antioxidant enzyme activities in ‘Süper domates’ genotypes harvested for two years in six different soil salinity areas in Iğdır University Agricultural Application and Research Center. In addition, Native Polyacrylamide Gel Electrophoresis (Native-PAGE) was performed separately on the seeds whose SOD, CAT and APX enzyme activities were determined spectrophotometrically during the study. After electrophoresis for each enzyme, the presence of enzymes was demonstrated by performing substrate staining on the gels. In the light of the data obtained, while the protein activity was low in the seeds harvested in saline alkaline soils, the highest protein activity was determined in the seeds taken from saline soils. SOD and CAT enzyme activities were found to be highest in salt stress caused by soil salinity in the plant and in salinealkaline areas, while SOD and CAT activities were calculated lower in soils harvested from normal soils. In the APX enzyme activities, it was observed that the activity increased from normal soil to saline-alkaline soil conditions. The results showed that the antioxidant activity in the harvested seeds increased as the severity of salt stress increased in the plant depending on the soil salinity and alkalinity.
