Abstract

Purpose: To determine the protective effect of alpha lipoic acid and octreotid against oxygen radicals in rabbit lens. Materials and Method: Rabbits weighing 2500-3000g were used and we divided them into 5 groups with randomly selected 5 rabbits. Rabbits exposed to 2J/cm2/h of ultraviolet A (UV A) radiation in the range 320-405 nm for 12 hr per day within 90 days. At this time group 1 (control group-G1) was not applied any procedure. Group 2 (UV A group-G2) only exposed to UV A radiation. Group 3 (PUVA group-G3) was applied 8-methoxypsoralen and UV A Group 4 (Alpha lipoic acid group-G4) was applied 8-methoxypsoralen+UV A+alfa lipoic acid. Group 5 (Octreotide group-G5) was applied 8-methoxypsoralen+UV A+octreotide. At the end of 90 days the rabbits were killed by decapitation and then eyes were enucleated. Both eyes of each rabbit were used for biochemical evaluation. Lens malondialdehyde (MDA), glutathione peroxidase (GSH-PX), superoxide dismutase (SOD) levels were measured at biochemical analysis Results: Biochemical analysis of each group indicated that lens MDA levels were lower in alpha lipoic acid group and octreotide group compared with UV A group (P<0.01) and PUVA group (p<0.001). Both lens SOD levels (p<0.01) and lens GSH-PX levels (p<0.001) were higher in alpha lipoic acid group and octreotide group compared with UV A group and PUVA group. SOD and MDA levels did not show statistical difference between control group and medical groups but there was statistical difference for GSH-PX levels (p<0.01). Lens SOD and GSH-PX levels were higher in control group Key Words: Ultraviolet A, Alpha lipoic acid, Octreotid, PUVA, Antioxidants compared with UVA and PUVA groups (p<0.01), and MDA levels were lower in control group compared with the same groups (p<0.05, p<0.01, respectively). Conclusion: We conclude that alfa lipoic acid and octreotid which are considered as free radical scavengers protect the eye from the damaging effect of UV exposure.

Özet

Amaç: Ultraviyole A+Psoralen tedavisi alan deneklerin lenslerinde alfa lipoik asit ve oktreotidin oksidatif hasara karşı koruyucu etkisini araştırmak. Gereç ve Yöntem: Denekler 2500-3000 gram ağırlığında tavşandan oluşturulup, her grupta 5'er adet olmak üzere 5 gruba ayrılmıştır. Deneklere 90 gün boyunca hergün 12 saat 320-405 nm 2J/cm2/sa ultraviyole A uygulanmıştır. İkinci gruba (UV A grubu-G2) sadece ultraviyole A uygulanmıştır. Üçüncü gruba (PUVA grubu-G3) 8-methoxypsoralen (8-MOP)+ultraviyole A uygulanmıştır. Dördüncü gruba (Alfa lipoik asit grubu-G4) 8-MOP +ultraviyole A+alfa lipoik asit uygulanmıştır. Beşinci gruba (Oktreotid grubu-G5) 8-MOP+ultraviyole A+oktreotid uygulanmıştır.
