Abstract

Objective: The aim of this study is to isolate placental fibroblasts from chorionic villous tissue, culture and characterize. Materials and Methods: For this purpose, placental fibroblasts were isolated from chorionic villous tissue of first trimester öf human placenta. In the first step, chorionic villous tissue was carefully dissected free of membranes and decidua under sterile conditions. In the second step, tissue fragments were trypsinized for 1 hour at 37aC by using 0,25% trypsin-EDTA solution and cell pellets were resuspended in DMEM/F12 and cells were incubated in 25 cm2 flask in humified (5% CO2 and 95% O2) air at 37QC. When the cells became confluent, they were passaged. After four passages, the placental fibroblasts were used for characterization tests. For this purpose, in the first step, cell growth was determined at four different initial cell seeding densities and growth curves were obtained. In the second step, plating efficiency and soft agarose test were did to investigate colony formation of placental fibroblasts were incubated in four different serum concentrations and growth curves were obtained. Results: Results showed that placental fibroblasts had a high growth capacity in Group I. Plating efficiency and soft agar tests were showed that there was no colony formation. And the results also showed that the best serum concentration to grow placental fibroblast cells is 20%. Conclusion: The results showed that placental fibroblasts had high growth capacity. But they hadn't got ability of colony formation under normal conditions. According to these results, placental fibroblasts can be used to prepare fibroblast feeder-layer.
