Abstract

AIM: The aim of this was to identify the effects of EGb76l and Pentoxifylline, which were both reported to prevent free radical formation on previous investigations, on rat hind limb skeletal muscle reperfusion injury. MA TERIAL AND METHODS: 84 male Spraque -Dawley rats were divided into twelve groups. (Control, control + EGb761, control + pentoxifylline, control +EGb761 + pentoxifylline, ischemia, ischemia + EGb76l ischemia+ pentoxifylline, ischemia + EGb76l + pentoxifylline, reperfusion, reperfusion + EGb76I, reperfusion+ pentoxifylline, reperfusion + EGb761 + pentoxifylline) All the tissues except femoral artery and vein of the hind limb were dissected in an attempt to prevent collateral circulation. Animals were subjected to 3.5 hours of ischemia followed by 1 hour reperfusion depending on the experimental group. EGb761 (50 mg/kg) or Pentoxifylline (50 mg/kg) or both were administred by intravenous route in 10 minutes. Wet /dry weight ratio, histopathologic findings A tissue malondialdehyde (MDA) level of the rectus femoris muscles were determined. RESULTS: Wet/dry weight ratio, histopathologic findings & tissue malondialdehyde (MDA) level of the reperfusion drug groups was found to be regressed down to the level of ischemia groups in respect to reperfusion group. CONCLUSION: Both drugs were found to be effective in preventing the skeletal muscle from reperfusion injury. Further clinical investigations are necessary for the clinical application of these findings.

Özet

AMAÇ: Bu çalışmada alt ekstremite iskelet kası reperfüzyon hasarının önlenmesinde serbest radikal giderici özellikleri daha önce çeşitli çalışmalarla gösterilmiş olan EGb761 ve pentoksifilinin etkilerinin araştırılması planlanmıştır. GEREÇ VE YÖNTEM: 84 adet Spraque-Dawley cinsi sıçan kullanılmıştır. Sıçanlar 12 gruba ayrılmıştır. (Kontrol, Kontrol + ECb761, Kontrol + Pentoksifilin, Kontrol + EGb761 + Pentoksifilin. İskemi, İskemi + EGb76l, İskemi + Pentoksifilin, İskemi + EGb76I + Pentoksifilin, Reperfüzyon, Reperfüzyon + EGb76l, Reperfüzyon + Pentoksifilin, Reperfüzyon + EGb76l + Pentoksifilin) Sıçanların alt ekstremitesinde femoral arter ve ven dışında bütün dokuların disseke edilmesini takiben femoral artere 3,5 saat süreyle mikrovasküler klemp konmuş, reperfüzyon gruplarında l saat reperfüzyon yapılmıştır. Her iki ilacın klinik araştırmalarda kullanılması umut verici olabilir.
